外周损伤对脊髓P物质免疫反应的影响及Mel1a受体在脊髓中的定位

【摘要】 1. P物质是一种神经肽,脊髓P物质主要在背根中与疼痛调节相关的小型初级感觉传入神经中表达,在中枢和周围神经系统起着重要作用。运用免疫组织化学和光密度测定法,观察中华大蟾蜍坐骨神经横断后脊髓中P物质的变化。在对照动物中,背外侧束中P物质免疫反应纤维密度最高,胞体内没有发现任何P物质免疫阳性标记。手术后,损伤侧背外侧束中的P物质免疫反应纤维减少。这种变化是在轴突损伤3天(0.84±0.02)后观察到的,并且在第8天(0.50±0.03)和第15天(0.45±0.04)持续出现,在第20天(0.60±0.04)、30天(0.69±0.03),手术侧和对照侧之间已经没有明显的区别,表明P物质表达的恢复。这些结果说明中华大蟾蜍可以用作研究周围损伤影响的模型,对阐明P物质在疼痛神经病患者中的作用会有所帮助。2.褪黑素是一种具有广泛生理功能的神经内分泌激素,通过与其受体的结合来实现对中枢神经系统的调节作用。为了进一步了解褪黑素对脊髓不同板层的调节差异,我们运用免疫荧光化学方法,观察了Mel1a受体在无蹼壁虎脊髓中的分布,并对阳性结果进行统计分析。结果显示I-V和X板层的Mel1a受体免疫阳性细胞胞体形体较小,形态多为圆形;VII层的阳性细胞形体较大,形态不规则,呈圆形、三角形或椭圆形三种。统计结果表明,VI层(22.36±2.36个/mm2)、VIII层(24.07±2.05个/mm2)和IX层(30.56±2.87个/mm2)较I-V层(450.25±9.65个/mm2)阳性细胞明显偏少(P<0.05);VII层(476.90±11.26个/mm2)和X(480.64±12.98个/mm2)板层阳性反应细胞密度同I-V板层(450.25±9.65个/mm2)相比没有显著性差异(P>0.05)。提示褪黑素通过其受体介导除了对脊髓的感觉传输具有调节作用以外,还可能参与调节爬行动物的运动功能。

【Abstract】 1. Substance P is a neuropeptide prominently expressed in small primary sensory afferents that is involved in the modulation of pain-related information in the spinal dorsal horn.Using immunohistochemistry and optical densitometry, substance P was investigated in Bufo bufo gargarizans spinal cord after sciatic nerve transection. In control animals, there was a high density of substance P fibers in the Lissauer’s tract. No substance P label was found in any cell bodies. After axotomy, substance P immunoreactive fibers decreased in the Lissauer’s tract on the same side of the lesion. The changes were observed at 3 days(0.84±0.02) following axonal injury and persisted at 8(0.50±0.03) and 15 days(0.45±0.04). At 20 days(0.60±0.04) and 30 days(0.69±0.03), there was no significant difference between the axotomized side and the control one, thus indicating a recovery of the substance P expression. These results indicate that Bufo bufo gargarizans may be used as a model to study the effects of peripheral axotomy, contributing to elucidate the Substance P actions in the pain neuropath.2. Melatonin is a kind of neuroendocrine hormone, which regulates the functions of Central Nervous System(CNS) by its receptors.In order to learn more about the regulatory function of melatonin to different lamina of spinal cord, we localized and characterized Mel1a receptor in the spinal cord of Gecko swinhoniss by immunofluorescence. Studies demonstrated the presence of Mel1a receptor located mainly in laminae I-V, lamina VII and lamina X of the spinal cord. The positive cells were small and mostly round in laminae I-V and X, while in lamina VII the positive cells were large and the shapes in them were cycloidal, triangle and elliptical. The statistical results showed that the positive cells in lamina VI(22.36±2.36/mm2), lamina VIII(24.07±2.05 /mm2) and lamina IX(30.56±2.87/mm2) were evidently lesser than those in laminae I-V(450.25±9.65/mm2)(P<0.05); The positive cells in lamina VII(476.90±11.26/mm2) and lamina X(480.64±12.98/mm2) did not have significant difference with those in laminae I-V(P>0.05). These results indicate that melatonin in spinal cord may play a role in the movement of reptile by its receptors besides the modulatory effect of melatonin on the mechanisms of sensory transmission of the spinal cord.
