几个牡丹品种的耐湿热生理生化特性研究

【摘要】 牡丹(Paeonia suffruticosa Andr.)是中国十大传统名花之一,其品种丰富,花型花色千变万化,深受广大民众喜爱。牡丹喜温暖及冷凉气候,持续高温高湿会对其造成伤害。牡丹在园林绿化美化中应用十分广泛,而南方地区夏季高温高湿使其观赏价值和应用范围受到影响。本试验以6个牡丹品种为材料,在不同温度(25℃、30℃、35℃、40℃)、不同土壤湿度(40%、60%、100%)、不同相对空气湿度(80%、90%、95%)条件下研究牡丹叶片生理生化指标变化,旨在为牡丹的耐湿热鉴定提供简单实用的方法,为牡丹的抗性育种、选种打下一定的基础,也为牡丹在高温高湿地区的推广应用提供更科学的理论依据。本试验主要结论如下:1.利用脯氨酸含量、蛋白质含量、叶绿素含量、丙二醛含量、相对含水量、电导率及超氧化物歧化酶、过氧化物酶、过氧化氢酶的活性等生理生化综合指标,鉴定了几个牡丹品种的耐湿、热性。结果表明,肉芙蓉、乌龙捧盛抗热性较强;藏娇次之,鲁荷红和脂红最弱;藏娇和脂红对高土壤相对含水量的抵抗能力较强,鲁荷红居中,胡红最弱;肉芙蓉比鲁荷红对高空气相对湿度的抵抗能力强。2.在35℃高温条件下,利用因子分析法和隶属函数法对供试材料的9个生理指标进行分析,得出了牡丹耐热性综合评价值(D),并依此进行了耐热性判别,建立了以游离脯氨酸含量(X1)、丙二醛含量(X2)、电导率(X4)、过氧化氢酶活性(X8)为自变量的耐热性预测回归方程:D=0.515-0.00054X1+0.000693X2+0.001X4-0.00069X8。3.在100%土壤相对含水量条件下,利用因子分析法和隶属函数法对供试材料的9个生理指标进行分析,得出了牡丹耐高土壤相对含水量能力综合评价值(D),并依此进行了耐高土壤相对含水量能力判别。建立了以游离脯氨酸含量(X1)、丙二醛含量(X2)、过氧化氢酶活性(X8)为自变量的耐高土壤相对含水量能力预测回归方程:D=1.947-0.00161X1-0.00132X2-0.00097X8。4.邓肯氏(Duncan)新复级差检验法和隶属函数综合分析显示,高温、高湿(含高空气相对湿度及高土壤相对含水量)胁迫下的脯氨酸含量、蛋白质含量、丙二醛含量、相对含水量、电导率及超氧化物歧化酶、过氧化氢酶的活性变化规律性强、差异性显著,可以作为鉴定的参考指标。其中游离脯氨酸含量、丙二醛含量、电导率、过氧化氢酶活性4个指标对牡丹耐热性鉴定效果较好;游离脯氨酸含量、丙二醛含量、过氧化氢酶活性3个指标对牡丹耐高土壤相对含水量能力的鉴定效果较好。

【Abstract】 Tree peony(Paeonia suffruticosa Andr.), a traditional flower in China, gets itspopularity for its rich cultivars and a wide range of colors. The cool climate is suitablefor it, but in cultivation high temperature and humidity would potentially make atremendous damage, and then there for their ornamental values are decreased andtheir appling region are limited. With the aim of providing simple and appliedmethods for the identification of tree peony’s heat and humidity tolerant, and offeringscientific basis for the application of peony in high temperature and high humidityarea, six cultivars of peony were used as materials under the different temperature(25℃, 30℃, 35℃, 40℃), the different SRWC (40%, 60%, 100%), the differentrelative air moisture (80%, 90%, 95%) in this study, and physiological andbiochemical changes under heat and humidity treatment were studied.The main results were as follows:Results of the leaf physiological and biochemical changes showed that:1. Physiological and biochemical indexs of proline, protein, chlorophyll,malondialdehyde, relative water content, conductivity and super oxide dismutase,peroxidase, catalase activity were used to evaluate the heat and humidity tolerantcharacters of peony. Paeonia suffruticosa ’Rou Furong’ and ’Wu Longpengsheng’was heat tolerant, ’Cang Jiao’ was next, Paeonia suffruticosa ’Lu Hehong’ and ’ZhiHong’ were the weakest; Paeonia suffruticosa ’Cang Jiao’ and ’Zhi Hong’ were highsoil SRWC tolerant, Paeonia suffruticosa ’Lu Hehong’ was in middle, but Paeoniasuffruticosa ’Hu Hong’ wasn’t. The air moisture tolerant of Paeonia suffruticosa ’RouFurong’ is stronger than Paeonia sufrruticosa ’Lu Hehong’.2. Nine physiological and biochemical indexes were measured by using principalcomponent analysis and subordinative function under the temperature of 35℃. The comprehensive value of tree peony’s heat tolerant was obtained and evaluated. Usingthe proline content (X1), the malondialdehyde content(X2), the conductivity(X4),catalase activity(X8) as variable established forecast regression function, the bestregressive function was founded to forecast other tree peony’s heat tolerant:(D=0.515-0.00054X1+0.000693X2+0.001X4-0.00069X8).3. Nine physiological and biochemical indexes were measured by using principalcomponent analysis and subordinative function under the soil SRWC of 100%. Thecomprehensive value of SRWC tolerant was obtained and evaluated. Using theproline content (X1), the malondialdehyde content(X2),catalase activity(X8) asvariable established forecast regression function, The best regressive function wasfounded to forecast other tree peony’s SRWC tolerant:(D=1.947-0.00161X1-0.00132X2-0.00097X8).4. Results of Duncan and subordinative function analysis showed that: solubleprotein, malondialdehyde(MDA), super oxide dismutase(SOD), Catalase(CAT),peroxidase(POD) activity and conductivity was strong to the change regularity andremarkable to difference under the heat and humidity tolerant (contain high airmoisture and soil SRWC). The effect of free proline, MDA content, conductivity,CAT activity was fairly good to identify heat tolerant of tree peony; and free prolinecontent, MDA contents, CAT activity were fairly good to identify the tree peonyhumidity tolerant.
