芽孢杆菌制剂对小鼠下丘脑—垂体—肾上腺—免疫器官的影响

【摘要】 目的:本实验研究了枯草芽孢杆菌Pab04和蜡样芽孢杆菌PAS38制剂对小鼠神经—内分泌—免疫系统的影响,为生产实践提供理论依据。方法:①将72只雄性健康小鼠随机分为芽孢杆菌PAb04组、芽孢杆菌PAS38组、抗生素组、空白组4组。芽孢杆菌PAb04组和芽孢杆菌PAS38组分别饲喂含活菌10~6cfu/g饲料的日粮。抗生素组饲喂含土霉素20μ/g饲料,空白组饲喂基础日粮。小鼠饲分别喂至20、40、60天后每组各宰杀6只,运用免疫组织化学SABC法,对小鼠的下丘脑、垂体、肾上腺、脾脏、胸腺中促肾上腺皮质激素(adrenocorticotropic hormone,ACTH)表达的检测,分析ACTH作为神经—内分泌—免疫轴上的重要的激素在神经免疫内分泌之间的相互作用、相互关系以及芽孢杆菌制剂对其生成分泌的影响。②运用免疫组织化学SABC法对免疫器官脾脏、胸腺中的CD4、CD8T淋巴细胞进行染色和检测,分析芽孢杆菌对小鼠免疫的影响及作用。③将96只小鼠随机分为芽孢杆菌PAb04组、芽孢杆菌PAS38组、抗生素组、空白组4组,运用MTT法,检测小鼠淋巴细胞转化能力。结果:①两株芽孢杆菌制剂对下丘脑、垂体、肾上腺、脾脏、胸腺组织中ACTH的表达,均有稳定的抑制作用,使ACTH的生成和分泌减少。结果表明芽孢杆菌制剂对小鼠神经—免疫—内分泌轴有一定的影响,对神经和内分泌进行温和调节,增强免疫有作用,使机体处于稳态。②芽孢杆菌制剂组对小鼠CD4、CD8的影响缓和有效,CD4/CD8的值趋于机体正常值2。Pab04组在20、40、60天脾脏CD4/CD8的变化分别是1.74±0.35,2.37±0.23,2.28±0.65;PAS38组在20、40、60天脾脏CD4/CD8的变化分别是2.17±0.49,2.63±0.41,2.55±0.56;抗生素组在20、40、60天脾脏CD4/CD8的变化分别是1.74±0.22,4.11±1.83,0.81±0.26;空白组在20、40、60天脾脏CD4/CD8的变化分别是1.26±0.31,0.94±0.42,2.26±0.56。抗生素组和空白组CD4/CD8波动大,对于机体建立稳态不利。③日粮中添加芽孢杆菌制剂的两组,淋巴细胞转化率显著高于抗生素组和空白组,说明,微生态制剂对机体的免疫有促进作用。

【Abstract】 Objective: This paper study the effect of Nervous-Endocrine-Immune System in rats by Additive of Gamma Germ. Method:①72 male rats were divided into 4 groups. Pab04 group was fed diet with additive of B. subtilis Pab04. PAS38 group was fed diet with additive of B. cereus PAS38. Antibiotics group was fed with Oxyteracycline. Control group was feed the basic diet. Immunohistochemistry SABC was used to measure ACTH of mouse hypothalamus, pituitarium, adrenal, splenic, thymus at the day of 20, 40, 60.②Immunohistochemistry SABC was used to measure CD4, CD8 of mouse splenic, thymus at the day of 20, 40, 60.③96 male rats were divided into 4 groups. MTT was used to measure the transformation of mouse splenic lymphocyte at the day of 14, 21, 28, 35, 42, 49, 56, 63.Conclusions:①The results indicated that Pab04 or PAS38 both have stably depressant effect on ACTH in mouse hypothalamus, pituitarium, adrenal, splenic, thymus, as well as reduce the production and secrete of ACTH. Microecology praeparation can reduced the contents of ACTH of tissue and enhance immunity.②Microecology praeparation has utility effective on rats CD4, CD8. CD4/CD8 has the tendency to normal value. The changes of splenic CD4/CD8 in Pab04 group mice are 1.74±0.35, 2.37±0.23, 2.28±0.65 during 60 days; PAS38 group mice are 2.17±0.49, 2.63±0.41, 2.55±0.56; antibiotics group mice are 1.74±0.22, 4.11±1.83, 0.81±0.26; control group mice are 1.26 ±0.31, 0.94±0.42, 2.26±0.56. Antibiotics and control group make big changes of CD4/CD8 in splenic during 60deys. It is disadvantaged for bodies to homeostasis.③The results indicated that Pab04 or PAS38 preparation improved the transformation of rat splenic lymphocyte respectively. This results were observe during th whole experiment that microecology preparation can improve the immune of rats.
