氢化可的松对人A375黑素瘤细胞株黑素合成的影响

【摘要】 目的:通过观察不同浓度的氢化可的松对人A375黑素瘤细胞的生长、黑素生成以及酪氨酸酶活性的影响,探讨氢化可的松治疗白癜风的作用机制。方法:用不同浓度的氢化可的松处理人A375黑素瘤细胞,通过MTT比色法观察氢化可的松对人A375黑素瘤细胞生长的影响,用NaOH裂解法测定黑素的含量,用多巴氧化法测定酪氨酸酶活性的变化。结果:1.氢化可的松浓度为10-7~10-5g/L时,随着药物浓度增高,细胞数目呈增加趋势;氢化可的松浓度为10-5~10-1g/L时,随着药物浓度增加,细胞生长呈下降趋势。与对照组相比,氢化可的松浓度为10-5 g/L时,细胞明显增殖,细胞体积增大;浓度为10-1时g/L,细胞生长明显受到抑制,细胞萎缩甚至变圆,细胞个数明显减少。2.氢化可的松浓度为10-7~10-3g/L时,随着药物浓度增高,黑素含量呈增加趋势,药物浓度为10-3g/L时黑素含量明显高于对照组;氢化可的松浓度为10-3~10-1g/L时,随着药物浓度增高,黑素含量呈现下降趋势,药物浓度为10-1g/L时黑素含量明显低于对照组。3.氢化可的松浓度为10-7~10-3g/L时,随着药物浓度增高,酪氨酸酶活性呈增强趋势,浓度为10-3g/L时酪氨酸酶活性明显强于对照组;浓度为10-3~10-1g/L时,随着药物浓度增加,人A375黑素瘤细胞酪氨酸酶活性呈下降趋势,药物浓度为10-1g/L时氨酸酶活性明显弱于对照组。结论::一定浓度的氢化可的松可以促进人A375黑素瘤细胞增殖,激活酪氨酸酶活性,促进黑素合成,为氢化可的松治疗白癜风提供了实验依据。
【Abstract】 Object: To observe the cell growth, melanogenesis and tyrosinase activity in A375 human melanoma cells after treatment with hydrocortisone of different concentration ;to study the mechanism of hydrocortisone in treating vitiligo.Methods: To treat A375 human melanoma cells in different concentration of hydrocortisone,to observe the growth of A375 human melanoma cells by MTT, to measure the content of melanin by spallation of NaOH, to measure the change of tyrosinase with dioxyphenylalanine oxidation.Results:1)When the concentration of hydrocortisone was between 10-7 g/L and 10-5g/L,it showed an increasing tendency of the amount of A375 human melanoma cells with the concentration of hydrocortisone raised, while the concentration of hydrocortisone was between 10-5 and 10-1g/L, it showed a decreasing tendency of the amount of A375 human melanoma cells with the concentration of hydrocortisone raised, compared with the control, when the concentration of hydrocortisone was at 10-5g/L , the cells proliferated obviously, the volume of the cells accreted; while he concentration of hydrocortisone was at 10-1g/L ,the growth of the cells was obviously inhibited ,the cells atrophied even turned circle, the amount of the cells decreased obviously.2)When the concentration of hydrocortisone was between 10-7and 10-3g/L, it showed an increasing tendency of the content of melanin when the concentration of hydrocortisone increased,when it was 10-3g/L, the content of melanin significantly increased comparing with the control; while the concentration was between 10-3g/L and 10-1g/L, the content of melanin got a tendency of decline with the concentration of hydrocortisone increasing; when it was 10-1g/L, the content of melanin significantly decreased comparing with the control.3)When the concentration of hydrocortisone was between 10-7and 10-3g/L,the activity of tyrosinase of A375 Human Melanoma Cells had an increasing tendency with the concentration of hydrocortisone increased, when it was 10-3g/L, the activity was obviously activated compared with the control; while the concentration was between 10-3g/L and 10-1g/L,the activity of tyrosinase of A375 Human Melanoma Cells showed a tendency of decline with the concentration of hydrocortisone increased, when it was 10-1g/L, the activity of tyrosinase was lowed obviously compared with the control.Conclusion: hydrocortisone of certain concentration may promote the growth of A375 Human Melanoma Cells, activate the activity of tyrosinase, enhance the production of melanin. And it provides an experimental evidence of treating vitiligo patients with hydrocortisone.
