开心散加减对血管性痴呆大鼠行为学和AChE mRNA的影响

Effects of Modified Kaixin Powder on Behavior and Acetylcholine Esterase mRNA in Vascular Dementia Rat

【摘要】 目的:开心散是古代治疗健忘的有效方剂,以往的实验研究表明该方能降低血液粘稠度,改善微循环,治疗老年性痴呆,故推测其具有减少或延缓血管性痴呆(VD)发生发展的作用。本研究在验证该方具有改善VD学习记忆功能的基础上,探讨该方对乙酰胆碱酯酶(AChE)mRNA表达水平的影响,为该方临床治疗VD提供理论支持,也为中西医结合临床防治VD奠定实验和理论支持。方法:应用永久性结扎双侧颈总动脉造成VD模型;30天后应用Morris水迷宫进行行为学测试;38天后应用RT-PCR半定量的方法检测大鼠海马AChE mRNA的表达水平;应用SPSS13.0软件对实验数据进行统计分析。结果:1.模型组大鼠第一天定向航行试验,7天定向航行试验,第8天空间探索试验,大鼠海马AChE mRNA表达等各项实验结果和空白组比较,P<0.05;2.与模型组相比,中西药合用组VD大鼠定向航行试验学习记忆潜伏期较短,空间探索试验大鼠在目标象限的游泳时间较长,在目标象限的路程比较大,目标区域的穿越次数较多,P<0.05;3.与模型组相比,他克林组VD大鼠定向航行试验学习记忆潜伏期较短,大鼠海马AChE mRNA表达水平较低,P<0.05;4.与模型组相比,开心散组和钩藤组空间探索试验大鼠在目标象限的游泳时间较长,目标象限的路程比比较大,P<0.05;5.中西药合用组、开心散组、钩藤组大鼠定向航行试验学习记忆潜伏期,空间探索试验目标象限的游泳时间,目标象限的路程比和目标区域的穿越次数与他克林组无统计学差异,P>0.05;6.中西药合用,开心散对VD大鼠海马AChE mRNA表达的影响与他克林无差异,P>0.05;结论:1.永久性结扎双侧颈总动脉造模可能引起VD大鼠GAPDH mRNA的过度表达;2.开心散、开心散加钩藤擅长改善VD大鼠的记忆能力;他克林擅长改善VD大鼠的学习能力;总体来说,开心散、开心散加钩藤改善VD大鼠学习记忆的能力与他克林相似,临床可辨证加减使用;3.开心散联用他克林具有改善VD大鼠学习记忆的作用,较单用他克林或者单用开心散效果好;4.他克林具有抑制VD大鼠海马AChE mRNA表达的作用,开心散、中西药合用抑制VD大鼠海马AChE mRNA表达的作用,与他克林相似。

【Abstract】 Objective: Kaixin Powder is an effective formula to treat morbid forgetfulness. Previousexperiments indicate it could degrade blood viscidity, improve microcirculation and cureAlzheimer’s Disease, so we deduce that it could prevent and cure Vascular Dementia. Studies onthe therapeutic effect of modified Kaixin Powders on Vascular Dementia rats and the effect ofmodified Kaixln Powder on acetylcholine esterase mRNA expression in hippocampus inVascular Dementia Rat supply the experiment foundation of Kaixin Powder and combinationTraditional Chinese Medicine and Westen Medicine on preventing and treating VascularDementia.Methods: The rat model of Vascular Dementia was established by ligating both commoncarotid arteries with silk thread. After 30-day-treatment, their learning and memory were testedwith Morris maze; 38 days later mRNA expression of AChE was measured with RT-PCRhalf-fix-quantify method; All the data was analyzed by SPSS 13.0.Results:1. The first day’s spatial navigation task latencies, 7 days’ spatial navigation task latencies,the results of probe task, and acetylcholine esterase mRNA expression in hippocampus of modelcontrol group rats were different from those of blank control group, P<0.05;2. Compared with model control group, combination of modified Kaixin Powder andTacrine group spatial navigation task latencies were shOrter, in probe task the swimming time,journey ratios and the times through platform zone in the target quadrant were bigger, P<0.05;3. Compared with model control group, Tacrine group spatial navigation task latencies wereshorter, the acetylcholine esterase mRNA expression was down-regulated, P<0.05;4. Compared with model control group, Kaixin Powder group and Ramulus Uncariae CumUncis group the swimming time and journey ratios were bigger in probe task, P<0.05;5. There was no significant difference between combination of modified Kaixin Powderand Tacrine group、Kaixin Powder group、Ramulus Uncariae Cum Uncis group and Tacrinegroup in spatial navigation task latencies, the swimming time, journey ratios and the times through platform zone in the target quadrant in probe task, P>0.05;6. There was no significant difference between combination of modified Kaixin Powderand Tacrine group、Kaixin Powder group and Tacrine group in acetylcholine esterase mRNAexpression, P>0.05.Conclusions:1. Two-vessel occlusion of the common carotid arteries maybe up-regulateglyceraldehyde-3-phosphate dehydrogenase mRNA expression in hippocampus of VascularDementia Rat;2. Modified Kaixin Powder, modified Kaixin Powder with Ramulus Uncariae Cum Unciscould be good at enhancing VD rats’ memory; Tacrine could be good at enhancing VD rats’learning; In a woed, modified Kaixin Powder, modified Kaixin Powder with Ramulus UncariaeCum Uncis could enhance VD rats’ learning and memory as well as Tacrine. We could use one ofthem acording to clinical need;3. Combination of modified Kaixin Powder and Tacrine could enhance VD rats’ learningand memory better than modified Kaixin Powder or Tacrine;4. Tacrine could down-regulate acetylcholine esterase mRNA expression in hippocampus ofVascular Dementia Rats, modified Kaixin Powder, combination of modified Kaixin Powderand Tacrine down-regulate acetylcholine esterase mRNA expression as well as Tacrine.
