兴安圆柏扦插繁殖技术及插穗生根生理基础的研究

【摘要】 本论文研究了适合兴安圆柏繁殖的育苗技术,研究外源IBA和NAA处理插穗基部对插穗中碳水化合物、蛋白质等物质的变化及保护酶系统、多酚氧化酶活性和内源激素水平的变化及其对生根的影响,试图为兴圆柏快速繁殖技术提供理论和技术支撑。(1)硬枝扦插是适合兴安圆柏无性繁殖技术的主要方式。以中河沙作基质,扦插苗质量最好,最长根可以达到14.3cm,且须根较多;在6月底或7月初选择两年生枝条的上部制穗,用IBA、NAA500mg/L处理扦插成活率可达80%以上。兴安圆柏嫩枝扦插比较困难,清水处理生根率低于10%,但是通过激素处理可以显著提高扦插成活率,最好的处理方式就是200mg/L的NAA,可使成活率提高到50%以上。(2)插穗内源激素的动态平衡,调控着不定根的发生。外源IBA和NAA处理插穗基部,提高了嫩枝插穗内源的IAA、IAA/ABA值和IAA/ZT值,降低了在ZT含量与IAAO活性。而激素处理同样提高了扦插初期硬枝插穗生根部位的IAA、IAA/ABA值,而降低了GA含量与IAAO活性,这些变化显然都有利于插穗的生根,提高了插穗的生根率。但是,所有的外源激素处理都不能降低插穗内ABA含量,并且硬枝生根部位的IAA、IAA/ABA、IAA/ZT变化规律相似。(3)在兴安圆柏生根过程中,SOD、POD、PPO活性表现不尽相同。外源激素显著提高了嫩枝的SOD活性,延缓了枝条的衰老,有利于嫩枝插穗的生根;插条体内的过氧化物酶(POD)、多酚氧化酶(PPO)活性呈现规律性的变化。在生根过程的不同时期,各酶的活性差异显著;表明兴安圆柏插穗的不定根发生和发展与POD、PPO、活性有密切联系。(4)IBA和NAA的施用使嫩枝插条内部水分含量、可溶性糖和可溶性蛋白在生根各阶段都处于一个较高的水平,而淀粉含量较低。说明激素处理促进了插穗内营养物质的转化,从而使插穗的生根率更高。本次试验对几种扦插方式进行了研究,通过比较得出了能够应用于生产中的兴安圆柏硬枝扦插技术,并对其生根机理进行了较为深入的探讨,有着明显的理论和实践意义。但对于嫩枝扦插技术的研究还需在激素配比、生根成苗机理方面进一步深入,使兴安圆柏的无性繁殖技术和理论体系更为系统,促进兴安圆柏优良种质资源的引种和快速繁殖。

【Abstract】 Sabina davurica (Pall.) Ant. was evergreen shrub species of Cupressaceae trees. They were recognized for their advantages of tolerance to wind and sand, drought and barren, etc. It can play a great role at the vegetation recovery in the desertfied areas. This paper carried out a research on the propagation techniques. That the external indolebutyric acid(IBA)and nathphylacetic acid(NAA)treat the bottom of soft wood,hard wood and can affected the variation of Carbohydrates, protein, protective Enzyme system, PPO activity. All of them try to provide the theoretical and realistic basement physiological mechanisms meaning.(1) The hard wood cutting was one rapid propagation techniques suit to Sabina davurica (Pall.) Ant. The root characters of the hard wood cutting was the best with the Fluviatile soil. Tbe longest root can reach 14.3cm and they have more little roots; The cuttings were made of the upper of the two years old hardwood branches in the end of June and the begin of July, and that survival rate was higher; Treated by NAA 500mg/L or IBA 500mg/L the survival rate was the highest. It was higher than 80%. Soft wood cutting for Sabina davurica (pall.) Ant.was more difficult, and the survival rate was lower than 10% treated by water. Using the suit technology increased the survival rate. It was significant differences in survival rate cutting treated by different hormone and concentration, the best survival rate was 50% when 200mg/L NAA was used to treat cutting.(2)The dynamic balances of endogenous hormone regulate adventitious rooting of the green wood cutting, the external indolebutyric acid(IBA)and nathphylacetic acid(NAA)treat could increase the contents of IAA, IAA/ABA and IAA/ZT and it decrease the contents of ZT and IAAO activity in soft wood cuttings during the rooting. The treat also increase the contents of IAA, IAA/ABA and IAA/ZT. It decrease the contents of GA and IAAO activity in hardwood cuttings during the period of root initiation. All the changes were good for rooting and they can increase the survival rate. But all treatments could not decrease the contents of ABA. The fluctuations of the IAA、IAA/ABA and IAA/ZT in the bottom of the hardwood cuttings were more similar.(3) There were so much difference during its rooting in SOD、POD and PPO. That the external IBA and NAA treated the cuttings can incarese the activity of SOD which delayed cuttage senescence whichwerebenefittorooting. Theactivitiesofperoxidase (POD) andpolyphenol oxidase (PPO) changed regularly. The differences of enzyme activities indifferent stages of rooting were very significant. It was indicate that the origin and development of adventitious roots in Sabina dawrica (Pall.) Ant. relative to POD, PPO, and IAAO. The differences of the promoting efficiency of IAA and IBA to different clones and differentoxidase activities ere significant or very significant.(4)The softwood cuttings were treated with indolebutyric acid(IBA)and nathphylacetic acid(NAA), the contents of soluble sugar、water and soluble protein is higher than water treatment during rooting. But the hormone treatment decreased the soluble amylum of the cuttings. It was showed that the translation of nutritive materials in cuttings could be promoted by the hormone treatment, moreover the rooting rate increased. Several cutting methods were studied, the technology of hard wood cutting for Sabina davurica (Pall.) Ant.were obtained, and the physiological mechanisms of rooting were researched further. These had great theoretical and realistic meanlng.But the proportion of hormone and the physiological mechanisms of rooting, forming sprout at soft wood cutting should be researched in-depth, in order to establish the mature clonal propagation technologic system and theory and promote the rapid propagation of Sabina davurica (Pall.) Ant..
