不对称含硅二烃基锡化合物的合成、结构表征及其生物活性

【摘要】 大量的研究表明,二烃基锡化合物具有一定的抗癌活性,不对称二烃基锡化合物由于含有两个不同的烃基具有更广泛的抗癌活性。在药物分子中,用硅原子对碳原子进行等电排置换后得到的药物称之为硅药,其特点是其原有药效可以保留,并有可能降低其毒性。为了系统地研究有机锡化合物的构效关系,本文以三甲基氯硅烷和三苯基氯化锡等为原料,合成了5个新的不对称含硅二烃基锡卤化物、9个新的不对称含硅二烃基锡二芳香酸酯和3个新的不对称含硅二烃基锡邻菲咯啉配合物。(1) R(R′Me2SiCH2)SnX-2 (2) Cy(Me3SiCH2)Sn(OCOAr)2 (3) R(R′Me2SiCH2)SnCl2(Phen) Ar=-C6H4-H,-C6H4-F-p,-C6H4-Cl-p,-C6H4-Br-p,-C6H4-NO2-p,-C6H4-OCH3-p,-C6H4-CH3-p,-C6H4-NO2-m,-C6H4-NH2-o R=Ph,Cy,Bu;X=Cl,Br;R′=Me,Et通过红外光谱、核磁共振谱、有机质谱等方法对目标化合物进行了结构表征。测试结果表明不对称含硅二烃基锡二芳香酸酯的中心锡原子与羰基上的氧配位,中心锡原子可能为六配位的扭曲八面体构型,二烃基锡邻菲咯啉配合物的中心锡原子与氮原子配位形成六配位的化合物。部分不对称含硅二烃基锡二芳香酸酯的生物活性测试结果表明,该类化合物对肺腺癌细胞(A549)具有较好的体外抑制率。本文对一系列新的具有生物活性不对称含硅二烃基锡化合物的初步研究,丰富了有机锡化学的内容,为研究开发安全高效的有机锡药物作了一些探索性的工作。
【Abstract】 Lots of research works showed that diorganotin compounds have obviousantitumour activity, and the activity of asymmetric diorganotin compounds is moreextensive because of two different alkyl groups. In drug molecule, whose certain Catom was substituted by a silicon atom was called silicon drug, the effect of the drugwill be reserved and the toxicity be reduced. In order to study systemically therelation between structure and effect of organotin, five diorganotin halidecompounds, nine Cyclohexyl(trimethylsilylmethyl)tin Arylcarboxylates and threediorganotin phenanthroline complexes containing silicon were synthesized in thispaper. (1) R(R’Me2SiCH2)SnX2 (2) Cy(Me3SiCH2)Sn(OCOAr)2 (3) R(R’Me2SiCH2)SnCl2(Phen) Ar=-C6H4-H,-C6H4-F-p, -C6H4-Cl-p, -C6H4-Br-p, -C6H4-NO2-p, -C6H4-OCH3-p, -C6H4-CH3-p,-C6H4-NO2-m, -C6H4-NH2-o R=Ph, Cy, Bu; X=Cl, Br; R’=Me, EtTheir structures were characterized by IR, 1H NMR, 13C NMR and MS. Theresults indicated that the Sn atom of Cyclohexyl(trimethylsilylmethyl)tinArylcarboxylates and diorganotin phenanthroline complexes containing silicon isprobably in a six-coordinated distorted octahedron.The preliminary bioassay tests showed that Cyclohexyl(trimethylsilylmethyl)tinArylcarboxylates have a good antitumour activity in vitro against A549 tumor cells.This preliminary research on a series of novel silicon-containing diorganotincompounds with biological activities may enrich the study of organotin chemistry,and provide scientific basis for the exploitation of high-safety and high-efficiencyorganotin pharmaceuticals.
