发酵法产邻苯二酚菌株的筛选及固定化细胞的条件优化

【摘要】 邻苯二酚是一种应用极为广泛的化学中间体,它所涉及的领域包括化学、农业、医学药品、轻工业、食品等,目前用于各个领域的邻苯二酚主要是依靠化学合成途径来生产,而近年来发酵法转化芳香烃化合物合成邻苯二酚日益引起人们的关注。从土壤中筛选到以苯甲酸钠为唯一碳源的产生邻苯二酚的菌株44株,其中以X16菌株产生邻苯二酚的能力最强。经形态特征及16S rDNA序列同源性比较,初步鉴定X16为假单胞菌Pseudomonasp sp.。经培养条件的初步优化,发酵液中邻苯二酚的含量达到1.0mg/mL;同时以海藻酸钠、明胶、聚乙烯醇以及海藻酸钠和聚乙烯醇的混合物对X16菌株进行细胞固定化研究,通过单一材料细胞固定和混合材料细胞固定的条件的初步优化,并对其固定效果的比较,发现海藻酸钠和聚乙烯醇混合的固定效果最好,发酵液中邻苯二酚的含量达到1.4mg/mL。提取X16的粗酶液进行检测其邻苯二酚1,2-双加氧酶和邻苯二酚2,3-双加氧酶降解邻苯二酚的活性,分别从pH、温度、金属离子及作用时间等方面对邻苯二酚1,2-双加氧酶和邻苯二酚2,3-双加氧酶进行初步研究,对今后研究邻苯二酚1,2-双加氧酶和邻苯二酚2,3-双加氧酶打下基础。
【Abstract】 Catechol is an important and applied extensive chemicalintermedia. The fields which is applied contain chemistry, agriculture,medicine, light industry and foodstuff etc. Although most of Catecholis produced by the way of chemo-synthesis, the means of transformingSodium benzoate to catechol by zymotechnics is more attractiverecently.Total 44 microorganisms were isolated from several soil samplesbased on sodium benzoate as a sole carbon source. The X16 has thehighest activity to producing catechol. The strain X16 was identifiedas Pseudomonasp sp. according to its character and 16S rDNA sequenceanalysis. The catechol production of Pseudomonasp sp. X16 was studied indetail. The output of catechol reach 1.0mg/mL after optimization of cultureconditions. At the same time sodium alginate, PVA, gelatin and sodium alginatemixed with PVA were used to the study of cell’s immobilization. To optimizated the condition and comparison immobilization effect of the single materials andthe mixing materials that the sodium alginate mixed with PVA was the bestmaterial, The output of catechol reach 1.4mg/mL.after ultrasonic disruption, measure the activity of catechol 1, 2-dioxygenaseand catechol 2, 3-dioxygenase in pH, temperature, metal ions and reaction timeetc. which built the foundation for the next research.
