苏木总酚定量方法、稳定性及提取工艺研究

【摘要】 苏木(Lignum Sappan)为豆科云实属植物苏木(Caesalpinia sappan L.)的干燥心材,又名苏方、苏方木、赤木、棕木、红柴等。为常用传统中药,其药理作用广泛,包括免疫抑制、抗肿瘤、镇静镇痛、降血糖、抗菌消炎、利尿及对循环系统的作用等。近年来药理研究发现苏木对心脏移植排斥反应具有较好的抑制作用。研究表明,苏木中总酚成分为其主要药效作用的有效部位。它包括巴西苏木素类、查耳酮类、原苏木素类及高异黄酮类衍生物等成分类型。为了配合对苏木抗心脏移植排斥反应的相关研究,本课题对苏木药材中的有效部位总酚性成分含量测定方法、稳定性及提取工艺进行了研究。本文首先以苏木精为标准物,建立了Folin-法测定苏木中总酚含量的分析方法。经方法学考察,用此法测定苏木中总酚的含量具有较好的稳定性和可行性;在建立了Folin-酚法测定苏木中总酚含量的基础上,以总酚含量为考察指标,对苏木中总酚的提取工艺进行了优化,确定了苏木的最佳提取工艺为A2B1C2D1,即加15倍量70%乙醇,加热回流提取两次,每次1小时;鉴于苏木总酚性成分的稳定性较差,本文又对苏木中的总酚性成分进行了稳定性研究。以总酚含量为考察指标,主要研究了光照和温度对稳定性的影响,结果表明光和温度对总酚含量影响均显著。研究结果对苏木有效部位、有效成分及制剂的提取和制备具有指导作用。本研究中,用Folin-法测定苏木中总酚含量、苏木提取工艺优化以及总酚稳定性研究至今未见报道,所得结果为苏木免疫抑制作用成分及其它活性成分的研究奠定了理论和应用基础。

【Abstract】 Lignum Sappan, the dried heartwood of Caesalpinia sappan L.,belongs to the family of Caesalpinaceae .It is also named for sappan, sappan wood, red wood, brown wood, red firewood and so on.It has been used in oriental medicine for a long time .Its pharmacology effect is broad, including immune inhibitory action , resisting the tumour effect , conquering the blood sugar effect , antibiosis the effect allaying inflammation , diuresis effect to the circulatory system effect , the calm analgesia effect, and so on.Pharmacology research discovers a logwood in recent years discriminating against a heart transplant reaction having stronger inhibitory action.Research indicates phenol , the general phenals is whose main pesticide effect material base.It includes brazilines,chalcones,dibezoxocin,derivatives,homoisoflavonoids and so on .To go with learning Lignum Sappan resist heart transplant rejective reaction,this text study and research the general phenol contents assay method,stabilitty,and handicraft of Lignum Sappan.In research, I first put the Heamatoxylin for standard thing, built Folin-law to determin the general phenals in Lignum Sappan , using this law admeasurement after study of method having the obvious stability.And process and result from the experiment are not bad reach,I optimize the extraction craft on the establishing the Folin-general law determin general phenal contents in lignum sappan ,by means of general phenol contents for consideration index.we have ascertained the best extractinghandicraft :A2B1C2D1,70% alcohol ,15 doubles , refluens extracting two times, every time 1 hour.In the end this text investigate the stability of the general phenol in lignum sppan.I study and research the illumination and temprature effect on stability.The result sign that the illuminition and temprature effect the stability significant.we must caution in the applying .The consequence guide the extraction and preparation of active part,active component and praeparatum of lignum sappan.In research, up to now,I first use Folin-phenol law to determin the general phenol contents,the extracting handicraft,and first research the stabiliyof genol phenol.The results establish rationale for immun-osuppressive action and active components researches.they had important significance for further making use of Caesalpinia Sappan L.
