HPLC-DAD指纹图谱技术用于峨眉味连品质评价研究

【摘要】 目的:建立峨眉产味连的指纹图谱,为峨眉产味连的GAP种植所必需的国际化质量标准提供技术依据。方法:采用超声波振荡提取等多种提取方法制备峨眉产味连特征性总提取物,以总生物碱含量为考察指标,选择出最佳提取方法;采用HPLC-DAD指纹图谱技术研究峨眉产味连指纹图谱,并进行色谱条件优化;在最佳的提取方法和色谱条件下,考察样品供试品溶液的稳定性、仪器精密度和重现性,并对所得谱图进行相似度分析;在最佳的提取方法和色谱条件下,进行样品和对照品检测,得到味连HPLC-DAD指纹图谱并进行特征峰分析;将峨眉不同乡镇产地所产味连进行指纹图谱相似度分析,考察其品质的一致性;将峨眉雅连指纹图谱与峨眉产味连指纹图谱进行比较,考察峨眉产味连指纹图谱的可靠性。结果:甲醇-盐酸超声波振荡提取为最佳提取方法,提取的味连总生物碱含量最高;用梯度洗脱方法能够得到各特征峰完全分离的指纹图谱;对味连指纹图谱的方法学考察表明,味连样品供试品溶液在48h内稳定,指纹图谱重现性好;峨眉不同乡镇产地味连的品质一致,峨眉产味连与雅连区别明显。结论:初步建立了峨眉产味连的HPLC-DAD指纹图谱,此指纹图谱可用于峨眉产味连的质量控制和峨眉规范化GAP种植基地生产的味连质量评价标准。

【Abstract】 Objective: To establish HPLC fingerprint for the identification of the Emei’s Coptis chinensis Franch.To offer the international quality standard which is needed for the planting of the Emei’s Coptis chinensis Franch.Methods: The ultrasonic-surge extraction and other extractions were applied to the preparation of the characteristic extraction of the Emei’s Coptis chinensis Franch.Take the content of the characteristic extraction as the guide line to choose the extraction. The HPLC-DAD method was applied to research the fingerprint of the Emei’s Coptis chinensis Franch and the conditions of chromatogram.At the best extraction and chromatogram conditions, we researched the stability of the sample solution, the precision of the HPLC,and the reproducibility.Then the comparability of the fingerprint of the Emei’s Coptis chinensis Franch was researched.At the best extraction and chromatogram conditions,the fingerprint of the Emei’s Coptis chinensis Franch was obtained after the sample solution and the reference substance solution had been tested.Then to analyze the characteristic peak of the fingerprint. Then to research the comparability of the fingerprints of the Emei’s Coptis chinensis Franch which yielded from different towns of Emei to study whether there quality was identical. The fingerprint of Emei’s Coptis chinensis Franch and Coptis dletoidea C.Y. Cheng et Hsiao were compared to study the reliability of the fingerprint of Emei’s Coptis chinensis Franch.Results: Methanol- HC1 ultrasonic-surge extraction is the best extraction,it’s content of total alkaloid of the Emei’s Coptis chinensis Franch is the most.The fingerprint which characteristic peak is completely separate can be obtained by means of gradient elution.After the research of the methodology of fingerprint ,it can be found that the sample solution of Emei’s Coptis chinensis Franch is steady within 48 hours,and the reproducibility of the fingerprint is wonderful.The quality of the Emei’s Coptis chinensis Franch which yielded from different towns of Emei is identical. The quality of Emei’s Coptis chinensis Franch is quite different with Coptis dletoidea C.Y. Cheng et Hsiao .Conclusion: The HPLC-DAD fingerprint of Emei’s Coptis chinensis Franch was established elementary,it could be used to research the quality of the Emei’s Coptis chinensis Franch and be used as the quality standard of the Emei’s Coptis chinensis Franch from the GAP base.
