九里香叶黄酮类成分的研究

【摘要】 九里香[Murraya exotical L.]为芸香科九里香属植物,是一种用途广泛的中药材,有行气止痛、活血散瘀、祛风除湿、麻醉镇惊功能。用于治疗胃痛、风湿痹痛,外治牙痛,跌扑肿痛,虫蛇咬伤。本实验利用水煎煮提取、大孔吸附树脂吸附、柱层析分离、有机溶剂重结晶等手段从九里香叶中得到了六种黄酮类成分,并利用IR,UV,MS,1DNMR, 2DNMR等现代光谱学手段辅以其他的定性方法鉴定出了五种成分的结构,它们分别是5,7,3′,4′′-四甲氧基黄酮(5,7,3′,4′′-tetramethoxyflavone)、5,7,3′,4′,5′-五甲氧基黄酮(5,7,3′,4′,5′-pentamethoxyflavone)、5,6,7,3′,4′,5′-六甲氧基黄酮(5,6,7,3′,4′,5′′-hexamethoxyflavone)、7-羟基-5,3′,4′-三甲氧基黄酮(7-hydroxy-5,3′,4′-trimethoxyflavone)和3′-羟基-5,7,4′-三甲氧基黄酮(3′-hydroxy-5,7,4′-trimethoxyflavone)。本实验还建立了5,7,3′,4′′-四甲氧基黄酮(5,7,3′,4′′-tetramethoxyflavone)、5,7,3′,4′,5′-五甲氧基黄酮(5,7,3′,4′,5′′-pentamethoxyflavone)这两种主要黄酮成分的HPLC含量测定方法,并且对购自不同地区的九里香茎叶进行了含量测定。本实验深入研究了九里香叶中的黄酮类化学成分,并填补了目前为止九里香药材没有质量标准的空白,为今后九里香的综合开发利用提供了新的理论基础和科学依据。
【Abstract】 Murraya exotica L., a well-known medicinal plant,which is widely used for treatment of stomachic diseases, inflammation, different body pains, trauma , bleeding caused by internal and external injury etc. It was been also used as a tonic and haemostatic agent. Due to the demand for its important medicinal use in China, it was largely cultivated in Yunnan and Guangxi Province.The alkaloids and terpenoids in this plant have been reported more before, but the flavonoids reported fewer. For the sake of the better use of this herb medicine, this paper describes the isolation, structural determinations and the analysis of the flavonids from the dried leaves of Murraya exotica L.Extraction and SeparationDried leaves (1kg) were extracted with water (15L, 12L, 10L, respectively) for three times (2h, 1.5h, 1h, respectively). The extracted solution was absorbed by macroporous resin, then eluented with 95%EtOH. The extraction was subjected to Al2O3 column chromatography using ethylacetate as mobile phase and silica gel column chromatography using ethylacetate-petroleumether as mobile phase to afford .JA(11.2g),JB(8.6g),JC(1.3mg),JD(104mg),JE(11mg),JF(8mg)。Compounds JA, JB, JC, JD, and JE was identified respectively by UV, IR, MS, 1DNMR and 2DNMR.as. 5,7,3′,4′-tetramethoxyflavone;5,7,3′,4′,5′-pentamethoxyflavone ;5,6,7,3′,4′,5′-hexamethoxyflavone;3′-hydroxy-5,7,4′-trimethoxyflavone;7-hydroxy-5,3′,4′-trimethoxyflavone .Compound JF is not identified now and hoped to be identified in the future .But for JA and JC,all the compounds were isolated from Murraya exotica L. for the first time.Analysis of flavonoids in the leaves of Murraya exotica L.In this paper we present a more sensitive quantification method by HPLC for analyzing the main flavonoids (JA and JB) in the leaves of Murraya exotica L.HPLC was performed on a ZORBAX C18 column(250 mm×4.6 mm,5μm)at 25℃with the rate of 1.2 ml/min ,using 58% methanol and 42% wateras mobile phase.The content of the two flavonoids-compounds JA and JB in the Murraya exotica L. getting from different regions were determined by the above method, the results showed that their contents were variety in different regions. The content of JA and JB in leaves is higher than in the stems. The content of JA and JB extracted getting from Yunnan province was 1.58% and 1.29%.While those getting from Hebei province only was 0.54% and 1.68%.
