人参皂苷Rg3的抗疲劳作用研究

【摘要】 人参皂苷Rg3是一种人参二醇类四环三萜皂苷,研究证明,人参皂苷Rg3具有抗肿瘤、舒张血管以及增强免疫力等作用,但其抗疲劳作用的报道甚少。本文通过小鼠负重游泳实验、爬杆实验和不负重游泳后测定小鼠肌、肝糖原含量、血尿素氮(BUN)含量、血清乳酸脱氢酶(LDH)活力、血乳酸(LA)含量以及肌肉组织ATP酶活力实验观察了人参皂苷Rg3的抗疲劳作用;并通过小鼠碳廓清实验和脾淋巴细胞转化等实验初步探讨其对免疫功能、外周血白细胞(WBC)、红细胞(RBC)、血红蛋白(Hb)、血清丙二醛(MDA)含量和血清总超氧化物歧化酶(SOD)活力的影响。结果表明,人参皂苷Rg3中、高剂量可明显延长小鼠负重游泳时间,提高疲劳小鼠的肝糖元含量;人参皂苷Rg3低、中、高剂量均可明显延长小鼠爬杆时间,提高疲劳小鼠的肌糖元含量,降低小鼠BUN含量,提高小鼠肌肉组织Na+-K+-ATP酶活力,降低小鼠游泳10min后血LA含量,证明人参皂苷Rg3有一定的抗疲劳作用。此外,人参皂苷Rg3中、高剂量可明显提高小鼠吞噬指数(k);人参皂苷Rg3高剂量亦能明显提高吞噬活性(α),降低疲劳小鼠的MDA含量,证明其具有一定的增强机体免疫功能和抗氧化损伤作用。

【Abstract】 In current society, along with the increasingly severe social competition and speeding up of living rhythm, fatigue has became more and more universal phenomenon and seriously impacts on physical strength and physical and mental health of human being. In addition, along with the rapid development of sports science, sports competition has been increasingly intense. Domestic and foreign scholars have focused on exercise capacity of competitors for a long time. Health products with reasonable and healthy nutrition security have been playing a significant role because of the effect on eliminating fatigue and physical restoration.The unti-tumor,vasodilatation, antiviral and immune function of ginsenoside Rg3 have been investigated. This experiment studies were on anti-fatigue effect of ginsenoside Rg3.1.Weight-loading swimming experiment and WBC,RBC and Hb content assayWe observed the weight-loading swimming time of mice through 5% weight-loading swimming experiment, and also assayed the content of WBC, RBC and Hb. The results showed that mid and high dosage of ginsenoside Rg3 could prolong weight-loading swimming time of mice, but had no effect on WBC, RBC and Hb.2.Pole climbing experiment and RES function experimentWe observed the pole climbing time of mice through pole climbingexperiment, and we also observed the function of RES of mice. Results demonstrated that three dosages of ginsenoside Rg3 could prolong pole climbing time of mice, mid and high dosage of ginsenoside Rg3 could increase the value of k and high dosage of ginsenoside Rg3 could increase the value ofα.3.The experiment on BUN content assay and LDH and muscle ATPase activity assayThrough assaying the content of BUN, the activity of LDH and muscle ATPase assay, we obtained the metabolism of nitrogen substance, the cleaning ability of LA and the enzyme activity of muscular in mice. The results showed that three dosages of ginsenoside Rg3 could decrease the content of BUN and increase the activity of Na+-K+-ATPase .4.Blood LA content assay experiment and splenic lymphocyte transformation function experimentThrough the assaying the content of blood LA at there time points, we obtained the accumulating and eliminating information of LA in mice. We also obtained SI. The results showed that three dosages of ginsenoside Rg3 could decrease the content of LA at after swimming 0min time point, but had no effect on the other two time points. Also, ginsenoside Rg3 had no function on splenic lymphocyte transformation.5. The experiment on muscle glycogen, liver glycogen and serum MDA content assay and activity of serum SOD assayWe observed the store information of muscle glycogen and liver glycogen in mice, through content of muscle glycogen and liver glycogenassay. At the same time, we obtained the unti-oxygen ability, through the content of serum MDA and the activity of serum SOD assay. Results demonstrate that mid and high dosage of Ginsenoside Rg3 could increase the content of muscle glycogen and liver glycogen and high dosage of ginsenoside Rg3 could decrease the content of serum MDA.The results of these studies have showed that compared with control ,ginsenoside Rg3 could prolong weight-loading swimming time and pole climbing time of mice, increase the value of k andα, the activity of Na+-K+-ATPase, the content of muscle glycogen and liver glycogen and decrease the content of BUN , LA and serum MDA of mice. The results have demonstrated that ginsenoside Rg3 has the effect on unti-fatigue, unti-oxygen and reinforce-immunity.
