生后早期音乐暴露对大鼠听皮层和前扣带回GluR2蛋白表达的影响

Early Music Exposure Modify the Expression Level of GluR2 Protein in Auditory Cortex and Anterior Cingulate Cortex of Rat

【摘要】 GluR2作为AMPA受体的重要组成部分,对Ca2+流具有明显的调节作用,在大脑兴奋性活动中起着重要的作用。研究其生后发育过程中的表达模式,以及环境、经验导致的表达变化,对揭示AMPA受体在中枢神经系统发育可塑性中的作用具有重要意义。本论文采用Western-Blotting技术,研究了大鼠听皮层和前扣带回AMPA受体GluR2亚单位蛋白的年龄-依赖性表达模式,以及音乐暴露对GluR2蛋白表达模式的影响。一、大鼠听皮层和前扣带回AMPA受体亚单位GluR2蛋白的年龄-依赖性表达模式应用Western-Blotting技术,在SD大鼠出生后不同的年龄段,检测GluR2的表达量变化。结果显示,随着年龄的增长,听皮层和前扣带回GluR2蛋白表达均呈现上调趋势。这种年龄-依赖性的上调表达模式可能与听皮层及前扣带回的功能发育相关联。二、生后早期音乐暴露对大鼠听皮层和前扣带回GluR2受体亚单位年龄-依赖性表达模式的影响在实验中,对大鼠采取不同的音乐暴露:音乐1和音乐2。音乐暴露自生后14天开始,分别在出生后28、42和56天检测听皮层和前扣带回GluR2的表达。结果显示,在经音乐2暴露后的各检测点均显示听皮层GluR2的上调表达;但经音乐1暴露后,表达水平与对照组相比没有显著变化。而在前扣带回,在出生后28天,两种音乐暴露均未表现出明显变化;但在42天龄时,影响开始显著;56天龄时,音乐2暴露组的GluR2表达与正常组仍存在差异性,但音乐1暴露组与正常组已不存在统计差异。提示某些音乐暴露能够改变大鼠听皮层和前扣带回的GluR2表达水平。

【Abstract】 GluR2, usually a main part of AMPA receptor, contributing to the regulation of Ca2+ influx, plays an important role in the brain excitatory activity. So it is meaningfully to study the developmental expression pattern of GluR2 proteins as well as the effects on the expression resulting from environments and experiences, which will reveal the roles of GluR2 and AMPA receptors in the developmental plasticity of CNS.Here we used Western Blotting method to the developmental expression pattern of GluR2 proteins in auditory cortex and anterior cinguiated cortex in SD rats. Also, we detected the changes of expression of GluR2 proteins in rats exposed to music. Our results provided evidences to reveal the mechanism of the plasticity of development in sensory function.1. Age-dependent expression pattern of AMPA receptors subunit GluR2 proteins in rat auditory cortex and anterior cinguiated cortexWestern blotting method was used to detect the expression changes of GluR2 at different postnatal time points. Results showed an increasing age-dependent expression of GluR2, which might be related to the functional development of auditory cortex and anterior cinguiated cortex.2. Effects of early music exposure on the age-dependent expression of GluR2 subunit proteins in auditory cortex and anterior cinguiated cortexRats were exposed to two kinds of music: Music1 and Music2, at postnatal days (PND) 28, 42, and 56 (from PND14, usually be thought as the beginning of ’critical period’)in auditory cortex (AC) and anterior cingulatecortex (ACC). Marked up-trend expression are presented in AC of the rats exposed to music2 at all age detected; and surprisingly, music1 exposed groups showed no significance by comparison with normal rats.In the ACC, we found remarkable discrepancy in expression of GluR2 between music exposed group and control group at PND42 and 56 between music2 group and control , but no statistical significance between the music1 group and control), while no obvious changes at PND28. Our survey indicated that exposure to music can modify the expression level of GluR2 protein in AC and ACC.
