黄芩药材指纹图谱及化学成分的测定

【摘要】 中药黄芩为唇形科黄芩属植物黄芩的干燥根,具有清热燥湿、泻火解毒、止血安胎等作用,是我国传统大宗药材之一。它具有多种药理作用,近年来国际上对黄芩的研究呈上升趋势。本论文分两个部分进行研究:第一部分介绍了黄芩的本草来源、化学成分、有效成分的含量测定,并对中药材HPLC指纹图谱、中药药效与无机元素的关系进行了简述。第二部分包括六个方面:1、对黄芩化学成分进行分离:提取和分离出黄芩苷并通过UV、IR、MS进行确证。2、对黄芩药材中的有效成分进行含量测定:通过紫外分光光度法测定药材中总黄酮含量,通过高效液相色谱法测定药材中黄芩苷含量,为工厂选择药材提供依据。3、对黄芩药材的提取工艺进行了研究:采用正交试验法优选黄芩药材的最佳提取工艺,并参考2005版药典进行了纯化,为工厂的工艺生产提供依据。3、对黄芩药材的指纹图谱进行研究:通过高效液相色谱法建立了不同产地黄芩药材的指纹图谱,为黄芩药材的质量控制提供依据。4、对黄芩药材的无机元素进行分析:采用电感偶合等离子体-质谱仪测定黄芩中的23种无机元素,有利于探讨中药药效与无机元素之间的关系。6、对黄芩指纹图谱共有峰化合物、药材提取物进行结构确证:采用液-质联用仪对指纹图谱9个共有峰化合物、药材提取物进行结构确证。

【Abstract】 Scutellaria baicalensis Georgi is one old and well-known drug in traditional Chinese medicines and widely used for treatments of bronchitis,hepatitis,tumors,and inflammatory diseases Recently, the international study Scutellaria more and more because of its various pharmacological effects.This paper includes two parts:The first part introduces the herbal medicine origin chemical components active ingredients determination,and give a brief description of Chinese herbal medicines HPLC fingerprint chromatogram the relationship between Chinese herbal medicines pharmacological effects and inorganic elements.The second part covers five aspects:1. Research the chemical components of Scutellaria: It separates baicalin and confirms the structures through the method of UV IR MS.2.Determinate the effective ingredients of Scutellaria:It determinates flavonoids by UV and baicalin by HPLC.This gives the factory a guide in choosing Chinese herbal medicines.3.Study the extraction process of Scutellaria: It selects the best one by Orthogonal Test and purificate flavonoids referring to the 2005 version Pharmacopoeia.This provides the factory with production basis .4.Research the fingerprint chromatogram of Scutellaria: It builds up the fingerprint chromatogram of different origin Scutellaria by HPLC.Yhis provides the basis for its quality control.5.Analyse inorganic elements of Scutellaria: It determinates 23 inorganic elements of Scutellaria by ICP-MS.This helps to explore the relationship between Chinese pharmacodynamics and inorganic elements.6.Confirmate the structures of the compounds of Scutellaria: Confirmate the structures of the compounds by LC-MS.
