海泡石固定化脱色菌及其对染料废水的处理研究

【摘要】 以海泡石作为一种新型的固定化载体,具有稳定性好、费用低、寿命长、传质阻力小、可循环利用等特点,本文研究了不同热活化温度对海泡石的结构特性和化学特性的影响,以及在去离子水和无机培养基中的基本性质,从结构和营养角度对其作为固定化载体的可行性进行了探讨,并讨论了各种影响固定化的因素。同时,还将固定化好的脱色菌应用到对染料废水的处理当中,以酸性红B为目标染料,讨论了海泡石活化温度、环境温度、pH值、不同海泡石的量、接种量、摇床转数、染料浓度、盐度等因素对染料对脱色反应的影响程度,还研究了对不同结构染料的降解情况,并运用紫外可见光谱法对降解过程和降解原理进行分析。实验结果表明,海泡石可以作为一种理想的固定化载体,固定化不仅提高了生物量,还增强了微生物对环境的适应力。活化温度为400℃的海泡石最适宜作为固定化载体用于微生物的固定化,海泡石对微生物的最佳固定化条件为:温度33℃,pH值为7~9,摇床转数为120r/min,载体结合时间为72h。对酸性红B脱色的最佳条件为:33℃、pH4~9、载体量为10g/L、接种量为2%、摇床转数为120r/min,最高脱色率可达94%以上。盐浓度对固定化B菌脱色率影响较小。对结构简单的单偶氮染料分子降解比较容易,而对相对复杂的双偶氮染料则要难降解。紫外可见光谱表明,B菌对酸性红B的降解,破坏了其结构中的共轭体系和芳香环结构。

【Abstract】 An experimental system was set up for immobilization by sepiolite, it had many features, such as excellent stability, low cost, long life, small mass transfer resistance, recyclable and so on. The paper studied on the structure and chemic features of sepiolite at different heat-treated temperatures, and the basic features in de-ionized water and inorganic culture medium. From structure and nutrition points of view, feasibility and every influence factor were discussed. Also, the immobilized decolorizing strain was supplied to dealing with waste water containing acid red B, the influence of activation temperature, circumstances temperature, pH value, the amount of carrier and microbe, the rate of shaker, dye concentration and salt degree on the strain decolorization were investigated, and the degradation of different structure dye was also studied. The process and principle of degradation were analyzed through UV — Vis spectrum.It was showed that sepiolite was able to be an optimum immobilization carrier. Not only the amount of microorganism in the system increased, but also the applicability of bacteria in different circumstances enhanced. The results also indicated that sepoilte heat-treated in air at 400℃ was the best immobilized carrier of the strain, and the suitable conditions for immobilized cells were determined as follows: temperature 33℃, pH7~9, 120r/min for the rate of shaker, 72h for combining. The optimum conditions for the dye decolorization of immobilized strain were temperature 33℃, pH4~9, the carrier amount of 10g/L, the microbe amount of 2% and the shaker speed of 120 r/min, the highest decolorizing rate could reach 94%, and the influence of salt degree was small. The decolorization of mono-azo dyes was easier than disazo dyes. UV — Vis spectra showed that the conjugate structure and aromatic part of acid red B were destroyed after degradation.
