野罂粟质量标准的研究

【摘要】 野罂粟(Papaver nudicaule L.)属于罂粟科罂粟属植物。野罂粟生物碱类物质具有镇痛、止泻、止咳的功效,作用与罂粟相似,而野罂粟所含罂粟碱为异喹啉类生物碱之一,其结构和药理作用均与鸦片作用成分菲类生物碱不同,没有麻醉性,而且不成瘾,是国家基本药物(分类号:4071078704),已经不列入麻醉药品管理范围。30多年来对野罂粟的研究日益深入。本文作者所在的实验室野罂粟所含生物碱的生物成分及其特有的镇痛、止咳平喘、止泻等药理作用的研究,先后得到了国家新药研究基金、国家自然科学基金、河北省自然科学基金和“国家1035”工程项目等多方面的基金资助;有关研究成果已经获得国家专利;野罂粟的新药研究也已经进入了临床研究阶段。所以制定野罂粟药材质量标准研究十分必要,本文通过对野罂粟中主要活性成分的定性和定量的系统研究,为建立野罂粟的质量标准提供了理论依据。全文由以下四部分组成:(一)综述分别从不同的方面对野罂粟研究进展进行了详细的概括和总结,展望了野罂粟的研究和开发前景;同时对开展野罂粟质量标准研究的目的、意义和必要性进行了阐述。(二)实验部分1、野罂粟质量标准的研究结合2005年版《中国药典》中有关药材质量标准的规定。2、野罂粟生物碱成分的指纹图谱研究采用HPLC方法,对野罂粟生物碱成分的指纹图谱的色谱条件进行了初步的研究。(三)、野罂粟与白山罂粟的形态学及化学成分比较为保障用药安全,本文分别从药材鉴别、显微鉴别、理化鉴别、HPLC色谱图研究四个方面对二者的区别加以研究。(四)结果与讨论以野罂粟碱为指标,选用样品浸泡过夜后回流提取的方法处理样品,建立了高效液相色谱测定方法,即以甲醇:0.1%三乙胺(55:45)为流动相,检测波长为258nm,对10份野罂粟样品中野罂粟碱的含量进行了测定。结果表明:野罂粟碱含量最高可达0.008965%,最低为0.000947%,其平均含量达0.002943%。所以,野罂粟中野罂粟碱的含量限度不低于0.002943%。该方法的线性关系良好(r=0.9996),回收率为96.5%,RSD为2.71%。该方法具有操作简便、快速、准确等优点。因此本文拟定了药典野罂粟碱含量测定草案。最后,本论文通过对野罂粟和白山罂粟的生药学特征、显微结构、生物碱类物质的HPLC图谱进行了对比研究,为这两种药物的鉴别和以后的开发利用提供了科学依据。
【Abstract】 The alkaloids is one of the important components in Papaver nudicaule L., such as amrensinine, re-framidine, nudicauline and nudicaulonol and so on, Nudicauline is found to be a good anodyne and will not cause addiction. On the development of the study on Papaver nudicaule L., these alkaloids in Papaver nudicaule L. have been shown, for example: anodyne, anti-chronic diarrhea and so on. It’s very important for studying and exploiting the alkaloids in Papaver nudicaule L.. To fulfill the mission of the People’Republilc of China PHarmacopoeia, that is to say the determination of the chemical of nudicauline in Papaver nudicaule L. will be added. this thesis includes four parts as follows:Review. A review of the alkaloids in Papaver nudicaule L. and the quality standard of Chinese traditional medicine were given.Experimental study1、 Studies on the quality standard for Papaver nudicaule L..A method for determination of liquiritin in Papaver nudicaule L. by HPLC was established.2、 Study on the fingerprint of the nudicauline in Papaver nudicaule L.. A method for the fingerprint of the nudicauline in Papaver nudicaule L. by HPLC was srudied. 3、 Compare morpHology, microscopic strucrure and HPLCchromatograms of Papaver nudicaule L. with Baishan Papavernudicaule. The comparison research on morpHology, microscopic strucrure and HPLC chromatograms for Papaver nudicaule L .and Baishan Pavernudicale is beneficial for discrimination of the two drugs. The result will provide foundation for the research on pHarmacology of the two drugs, and will promote exploitation and utilization of the two drugs.Result and discussion The mobile pHase was consisted of methanol-water-ethylamine(550:450:0.45) .and the detection wavelength was at 256nm. Then the content of nudicauline in ten samples was determined by this method. The result showed the content of nudicauline was between 0.001%~0. 008965%, the linear correlation was good (r=0.9996) and the average recovery was 96.5%. This method was simple reproducible and rapid, which was suitable for determination of nudicauline in Papaver nudicaule L..The original plant identification, macroscopical identification, microscopical identification and pHysical and chemical identification between Papaver nudicaule L. and Baishan Papavernudicaule were observed.
