直立白薇活性成分及质量控制研究

【摘要】 直立白薇Cynanchum atratum Bunge为萝藦科Asclepiadaceae鹅绒藤属Cynanchum Bunge植物,生于山地灌木丛中,在全国各地广有分布,特别是在东北三省,云南,贵州等地。具有清热凉血、利尿通淋、解毒疗疮的功效,临床上常用于温邪伤营发热、阴虚发热、骨蒸劳热、产后血虚发热、热淋、血淋、痈疽肿毒等症。现代药理研究表明,直立白薇的粗提物具有抗炎、平喘等生理活性。本文通过药理活性跟踪指导分离的方法,确定了直立白薇中具有抗炎镇痛作用的活性部位,并对其进行了重点分离。通过溶剂提取、萃取、AB-8树脂柱层析、HP-20树脂柱层析、硅胶层析、Sephadex LH-20柱层析,RP-18低压柱分离,RP-18中压柱分离以及C18柱半制备,从中分离得到了32个化合物,并运用UV、IR、MS、1H-NMR、13C-NMR、DEPT、HMBC、HMQC、COSY、TOCSY、NOESY等光谱技术和物理、化学方法鉴定了其中24个化合物的化学结构,其结构分别为BW-1:β-谷甾醇BW-2:β-胡萝卜苷BW-3:豆甾醇BW-4:壬二酸BW-5: 4-羟基苯乙酮BW-6: 2,4-二羟基苯乙酮BW-7:棕榈酸BW-8:正十八烷酸BW-9: 4-羟基苯甲醇BW-10:苯甲酸BW-11:β-香树素乙酸酯BW-12: 2,6-二羟基苯乙酮BW-13:正三十二烷醇BW-14: cynatratoside C BW-15: cynatratoside D

【Abstract】 Cynanchum atratum Bunge, locally called‘paiwei’(Asclepiadaceae), as one species of traditional Chinese medicine, widely distributs all around China especially in North China, Yunnan, Guizhou provinces. Cynanchum atratum Bunge has been used as a medicine for the treatment of hectic fevers ,acute urinary infection and abscesses for a long time in China. Morden research on pharmacology have indicated that the extracts of Cynanchum atratum have the activities of anti-inflammatory, analgesic activity, anti-asthma et al.In order to study further the resource of“Paiwei”, to illustrate the mechanism of the anti-inflammatory and analgesic activities and to find more active compounds, we use the bioactivity-screening on anti-inflammatory and analgesic activities to determinate the active fractions, and then to make the emphases isolation of this fraction. We are conducting a thorough solvent extraction, using non-polar macroreticular resin AB-8、HP-20、sephadex LH-20、ODS、HPLC、MPLC、silica gel chromatography and semi-preparative columns, we isolated 32 compounds from the chloroform fraction and 24 structures of them were established on the basis of spectral data including UV、IR、MS、1H-NMR、13C-NMR、DEPT、HMBC、HMQC、COSY、TOCSY、NOESY , physical and chemical properties, as follows: BW-1:β- sitosterol BW-2:β- daucosterol BW-3: stigmasterol BW-4: azelaic acid BW-5: 4-hydroxyacetophenone BW-6: 2, 4-dihyroxyacetophenone BW-7: palmitic acid BW-8: n-octadecily acid BW-9: 4-hydroxybenzenemethanol BW-10: benzoic acid
