利用废弃物作替代培养基制备微生物絮凝剂的研究

【摘要】 本论文的研究重点是利用工业废弃物作替代培养基制备微生物絮凝剂,并对培养条件优化、絮凝性能研究、絮凝剂的应用以及利用自制的放大反应器进行絮凝剂产生菌的扩大培养等方面进行了较为系统深入地研究与探讨。所得结论如下: 酱油曲霉在味精废水培养基和米面废水培养基中生长良好,絮凝剂分泌旺盛。其最佳培养条件分别为——味精废水浓度(CODCr)7300~7500mg/L,蔗糖1g/L,初始pH值为4;米面废水浓度(CODCr)1971~2138mg/L,蔗糖3g/L,NaNO30.3g/L,初始pH值为5;菌的絮凝剂分泌性与菌的生长有较好的相关性。 利用自制的放大反应器扩大培养酱油曲霉,可制备出高效微生物絮凝剂,对高岭土悬浊液的絮凝率最大可达99%以上。利用合成培养基、味精废水和米面废水培养酱油曲霉,可分别获得3.5g/L、4.0g/L、3.8g/L微生物絮凝剂。 絮凝实验表明:酱油曲霉产生的絮凝剂处理高岭土悬浊液时,絮凝剂投加量少,不会影响水质;在酸性或碱性的条件下,絮凝能力比较强;阳离子对絮凝剂的絮凝有明显促进作用,但价态不同,对絮凝的影响不同。 实际应用研究表明,酱油曲霉产生的絮凝剂对染料废水、啤酒废水、生活污水、河涌废水、乳品废水等多种实际废水均具有良好的净化效果,且三种不同培养基培养菌产生的絮凝剂对实际废水处理效果接近,其中对染料废水净化效果最好,对其进行处理时,加入高岭土联合使用,可加速絮体沉淀,CODCr、色度、浊度的去除率分别为72.7%、97.6%、96.5%。 酱油曲霉所产絮凝剂对Cr(Ⅵ)具有良好的还原作用,处理低浓度Cr(Ⅵ)溶液时还原率在90%以上。初步推断Cr(Ⅵ)的还原主要依赖于酱油曲霉体内合成并分泌到细胞外的溶解性还原酶。
【Abstract】 The emphases of the study was using industrial offal as substitutive media for flocculant-producing strains growing and producing bioflocculant. The optimization of culture condition for flocculant production, flocculating efficiency, and flocculant application were carried out and discussed systematically, the scale-up cultivation of the flocculant-producing strain in the self made reactor was also studied. The results were as follows:Aspergillus sojae could be cultivated in glutamic acid wastewater , flour wastewater, and excrete abundant bioflocculant. The optimum cultivating conditions were—glutamic acid wastewater concentration (CODCr) 7 300~7 500 mg/L, sucrose 1 g/L, the optimum pH was 4; flour wastewater concentration (CODCr) 1 971 ~2 138 mg/L, sucrose 3 g/L, NaNO3 0.3 g/L, the optimum pH was 5; The production of bioflocculant produced from the strain was parallel to the cell growth.Scale-up cultivating the Aspergillus sojae in the self made reactor could produce high-yield bioflocculant. When dealing with kaolin clay suspension, the flocculation ratio of the bioflocculant was over 99%. Cultivating the Aspergillus sojae in synthesize medium, glutamic acid wastewater and flour wastewater, the yield of bioflocculants was: 3.5 g/L, 4.0 g/L, 3.8 g/L, respectively.The flocculating experiment showed that when dealing with kaolin clay suspension, the addition of flocculants produced from Aspergillus sojae was small and did not influence the wastewater quality; The suitable pH value was acidity or alkaline; Metal cation could facilitate the flocculation activity of bioflocculant produced from Aspergillus sojae, but the facilitating effect was different because of the different charge of the cation.The wastewater purification experiments demonstrated that the bioflocculant had good purifying effect in the treatment of various wastewaters, and the purifying effect of bioflocculants prodruced by using different media was more or less the same. Especially in the treatment of yarn-dyed wastewater, when add kaolin to the wastewater, the sedimentation velocity was accelerated, and the satisfactory effect was achieved with the removal ratios of CODCr, chroma and turbidity were 72.7%, 97.6%, 96.5%, respectively.
