生后早期经验、活动改变大鼠听皮层NMDA受体NR2B亚单位蛋白表达
Early Experience and Activity Modifies Expression of NMDA Receptor Subunit NR2B Protein in the Rat Auditory Cortex

【摘要】 一、出生后早期听觉剥夺、活动对大鼠听皮层NMDA受体NR2B蛋白表达的双向调节 应用Western blotting检测技术,研究生后早期听觉剥夺、经验对大鼠听皮层NMDA受体NR2B蛋白表达的影响。结果表明,听觉剥夺使生后14天龄和28天龄动物听皮层NR2B蛋白表达水平明显下降(P<0.05、P<0.01),听觉剥夺7天后再给予纯音暴露则又使NR2B表达水平明显提高(P<0.01),呈现双向调节趋势。听觉剥夺和纯音暴露对生后42天龄大鼠听皮层NR2B表达不再产生明显调节作用(P>0.05)。结果提示,大鼠生后听觉发育关键期,听觉剥夺、经验可改变听皮层NMDA受体NR2B蛋白表达水平。研究结果为研究感觉功能发育可塑性的机制提供重要实验资料。 二、生后早期听觉经验对大鼠听皮层NMDA受体NR2B亚单位蛋白表达模式的影响 应用Western blotting和免疫组织化学技术,分别在生后14、28和42天龄大鼠上研究生后早期经验、活动对听皮层NMDA受体NR2B亚单位蛋白表达模式的影响。实验分为听觉剥夺组、纯音暴露组和听觉剥夺+纯音暴露组。结果表明,听觉剥夺组动物NR2B蛋白表达较对照组下降,其中14天龄组动物下降幅
【Abstract】 1. Bidirectional modulation of NMDA receptor subunit NR2B protein in the rat auditory cortex by Auditory deprivation and activity during early postnatal daysApplying Western blotting detection technology, we studied the influence of auditory deprivation and tone exposure to the relative level of NMDA receptor subunit NR2B protein expression of auditory cortex of SD rats during early postnatal days. The results showed that the NR2B protein expression of PND 14 and PND28 group rats reduced markedly (P<0.05, P<0.01). Tone exposure after auditory deprivation for 7 days made NR2B protein level increased significantly (P<0.05). It showed bidirectional adjustment trend. Auditory deprivation and tone exposure no longer showed obvious effect to NR2B protein expression of auditory cortex of PND42 rats(P>0.05).The results indicated that, during the critical period of rat postnatal auditory development, auditory deprivation and experience can alter the protein expression level of NMDA receptor subunit NR2B. The findings of this experiment provided important data to the study of the mechanisms for the developmental plasticity of sensory functions.2.
