类细菌素产生菌的选育及其发酵产物性质的初步研究

【摘要】 从四川某浓香型大曲酒酿造企业窖池里的黄水中,通过厌氧分离技术、纸片法初筛、三角瓶扩大培养、杯碟法复筛,得到一株代谢产物对革兰氏阳性致病菌、革兰氏阴性致病菌及部分真菌和酵母菌具有抑制作用的乳酸菌,菌株编号CF10。经形态观察、生理生化及分子生物学实验,鉴定该菌为布氏乳杆菌(Lactobacillus buchneri)。在排除酸性末端产物和过氧化氢的干扰作用后,CF10发酵上清液仍具有明显的广谱抑菌作用,故将这一抑菌活性成分纳入类细菌素的范畴。 在类细菌素高产菌株的选育过程中,采用紫外线(UV)与硫酸二乙酯(DES)多轮复合诱变,类细菌素高产菌发酵液相对效价较未诱变前提高了258%且具有良好的传代稳定性。 通过单因素分析和正交试验法,对目的菌CF10的培养基组成和发酵条件进行了探讨,确定了CF10的最佳培养基配方为:以2%麦芽糖为碳源,1%大豆蛋白胨加0.5%酵母粉为氮源,有微量K+、Mg2+、NH4+、Mn2+等金属离子存在。最适发酵条件为:培养温度37℃,起始

【Abstract】 A novel bacteriocin-like substance producing strain was screened from yellow-liquor samples by anaerobic isolation and scrip method and cylinder-dish method and was named CF10. The supernatant of CF10 culture have strong bacteriostasis activity against the gram-positive pathogenic bacteria and the gram-negative pathogenic bacteria and some fungi and microzyme. The strain was identified as Lactobacillus buchneri by morphologic characteristics and physiological-biochemical testing and molecular-biological testing. Excluded the interference of organic acids and hydrogen peroxide, strong bacteriostasis activity was also observed, we belong it to bacteriocin-like substance.In the process of mutation breeding bacteriocin-like substance high-producing strains, CF10 was treated with multiple times compound mutation by ultraviolet (UV) radiation and diethyl sulphate (DES), the relative potency of supernatant had increased 258% and it have stable inheritance.
