柱前紫外衍生—高效液相色谱法同时测定血清和饮料中18种游离氨基酸
Determination of 18 Free Amino Acids in Serum and Drinks by High Performance Liquid Chromatography after Ultraviolet Derivatization with 2,4-Dinitrochlorobenzene

【摘要】 目的:建立血清和饮料(葡萄酒、啤酒、葡萄汁、橙汁)中18种游离氨基酸(天冬氨酸、谷氨酸、组氨酸、丝氨酸、精氨酸、甘氨酸、苏氨酸、脯氨酸、丙氨酸、缬氨酸、甲硫氨酸、胱氨酸、异亮氨酸、亮氨酸、色氨酸、苯丙氨酸、赖氨酸、酪氨酸)的反相高效液相色谱分析方法。 方法:血清样品经偏磷酸沉淀蛋白,饮料样品经高速离心后,在碱性条件(pH9.0)下游离氨基酸与2,4-二硝基氯苯在90℃水浴中避光反应1.5h,生成具有紫外吸收的衍生产物,用反相C18柱分离,紫外检测器检测,内标法或外标法定量。分析条件:色谱柱:Phenomenex(USA),C18(ODS)柱,5μm,250×4.6mm;柱温为40℃;流动相A为乙腈,B为乙酸-乙酸钠缓冲液(pH5.20);梯度洗脱;流速1.0ml/min;检测波长360nm。 结果:用内标法或外标法测定18种氨基酸的标准曲线的相关系数范围均为0.9951~0.9999;相对标准偏差(RSD)为1.39%~4.67%;血样的检出限为0.20~7.72μg/ml,饮料样品的检出限为0.16~5.86μg/ml;血清加标回收率为65.9%~118%,饮料加标回收率为56.3%~125%。 结论:应用建立的方法成功地测定了血清、葡萄酒、啤酒、橙汁、葡萄汁等30份样品中的游离氨基酸。本法具有快速简便、准确度灵敏度较高、重现性好的特点,适合于血清和多种饮料中游离氨基酸的测定。
【Abstract】 Purpose: To establish a method of determination of 18 free amino acids[aspartic acid(Asp), alutamic acid (Glu), histidine (His), serine(Ser), arginine(Arg), glycine(Gly), threonine(Thr), proline (Pro), alanine (Ala), valine(Val), methionine (Met), cystine (Cys), isoleucine (He), leucine (Leu), tryptophane (Trp), phenylalanine (Phe), lysine (Lys), tyrosine (Tyr)] in serum and drinks (wine, beer, grape juice and orange juice) by pre-column derivatization-high performance liquid chromatography. Method: After removing protein in serum with HPO3 and removing precipitate in drink by centrifuging, the free amino acids in supernatant were derivatizated with 2,4-dinitrochlorobenzene in alkalescent media (pH9.0) in a water bath of 90℃. Then the derivatives were separated by C18 column and detected by UV detector, and the amount of amino acids was quantified with internal or external standard method. Analytical column: phenomenex (USA), C18(ODS) , 5μm, 250 × 4.6mm; column temperature:40℃; mobile phase A: acetonitrile, mobile phase B:HAc-NaAc (pH5.20); gradient elution with flow rate of 1.00ml/min; detection wavelength: 360nm.Results: The correlation coefficients of 18 free amino acids’ regression equations detecting by internal or external standard method were 0.9951~ 0.9999; the range of RSD was 1.39%~4.67%; the detection limits of serum samples were: 0.20~7.72μg/ml,the detection limits of drink samples were 0.16~5.86μg/ml; the recoveries for serum samples were 65.9%~118% and for
