STR基因座群体遗传学的研究及数据分析软件的开发与应用

【摘要】 目的 研究D15S659和D9S925基因座的群体遗传学和等位基因序列特征,积累群体遗传学数据,探讨其法医学应用价值,为法医遗传学提供新的候选遗传标记。 方法 以109名华东地区汉族无血缘关系个体的静脉血为研究样本。用Chelex法提取DNA,对D15S659和D9S925基因座进行PCR扩增;非变性聚丙烯酰胺凝胶连续缓冲系统垂直电泳进行分型。利用Modified-Powerstats标准版软件对分型结果进行统计分析,获得各基因座的等位基因频率、基因型计数及法医群体遗传学数据。 结果 D15S659和D9S925基因座在华东汉族群体中等位基因数分别为8和6;杂合度分别为0.826和0.798;个人识别率分别为0.944和0.894;非父排除率分别为0.648和0.596。 结论 在我国华东地区汉族人群中,D15S659和D9S925基因座等位基因频率分布较好,提供的多态信息量大。具有较高的非父排除率和个人识别能力,适合于法医学应用,是群体遗传学研究和法科学应用较理想的候选STR基因座。

【Abstract】 Objective To develop a set of new markers for forensic applications, D15S659 and D9S925 loci were chosen in present study. The polymorphism and allelic sequences of these loci were analyzed, and their forensic applications in Han population of eastern China are evaluated.Methods The EDTA-blood samples were collected from 109 unrelated individuals in Han population of eastern China. DNA was extracted with Chelex method. Each DNA sample was amplified using PCR for D15S659 and D9S925 loci. The PCR products were analyzed by non-denaturing vertical polyacrylaminde gel electrophoresis with continuous buffer system. The genetic and forensic parameters for the two STR loci were calculated by the Modified-Powerstats software.Results The allelic number of D15S659 and D9S925 was 8 and 6 respectively. The observed heterozygosity (H_o) of D15S659 was 0.826 and that of D9S925 was 0.798. The discrimination power (DP) of D15S659 was 0.944 and that of D9S925 was 0.894. The exclusion power (PE) of D15S659 and D9S925 were 0.648 and 0.596 respectively.Conclusion The distributions of allelic frequencies at two loci are satisfying in population studied. Present data suggest that D15S659 and D9S925 loci can be applied as new candidate markers for forensic individual identification and paternity testing.
