高压氧对新生大鼠缺氧缺血性脑损伤海马区细胞凋亡及NF-κB表达的影响

【摘要】 目的 1.研究高压氧(HBO)对新生大鼠缺氧缺血性脑损伤(HIBD)海马区神经细胞凋亡及NF-κB表达的影响。2.初步探讨HBO对缺氧缺血性脑损伤神经细胞的保护作用机制。3.观察NF-κB的活化在抗神经细胞凋亡中的作用。 方法 新生7日龄Sprague-Dawley(SD)大鼠72只,体重12-19g。将动物按随机表随机分为三组:1、假手术组(24只):游离左侧颈总动脉后不结扎,直接缝合皮下组织及皮肤,不进行缺氧处理;2、缺氧缺血(HIBD)组(24只):建立HIBD模型3、高压氧(HBO)组(24只):建立HIBD模型后,6h给予高压氧治疗一次,并在缺血缺氧后24h、48h、72h时各重复一次。假手术组、HIBD组和HBO组再分别于6h、24h、48h、72h各处死6只,灌注固定后取脑组织常规石腊包埋,切片,HE染色观察神经细胞坏死,采用TUNEL方法检测神经细胞凋亡,免疫组组织化学染色化法(SABC)检测NF-κB p65在海马区的表达。采用Image Pro Plus5.0专业图像分析软件进行图像分析,测量NF-κB p65表达阳性细胞数及凋亡阳性细

【Abstract】 Objective1.To study the effect of hyperbaric oxygenation(HBO) on NF-κB expression and apoptosis of neonatal rat hippocampal cell during hypoxia-ischemic brain damage(HIBD); 2. To observe the correlation-ship between NF-κB expression and apoptosis; 3. To investigate the mechanism of HBO neuronal protective effect after HIBD.MethodsSeventy-two 7-day postnatal Sprague-Dawley(SD) rats were randomly divided into 3 groups:(1)control group (n=24), (2)hypoxic-ischemic brain damage group (n=24), (3)HBO-treated group (n=24).In the HBO-treated group, rats received hyperbaric oxygenation at 6h,24h,48h and 72h after HIBD. The control group was not administrated. The expression of NF-kB in the hippocampal was measured by Immunohistochemical method. The apoptosis of the hippocampal cell was detected by terminal deoxynucleotidyltransferase-mediated 2-deoxyuridine 5-triphosphate-biotin nick end labeling (TUNEL) straining.
