小蓟的炮制工艺及质量标准规范化研究

【摘要】 论文对临床常用中药小蓟的炮制历史沿革、现代文献、有效活性物质基础、饮片规范化炮制工艺、质量标准及指纹图谱进行了较为系统深入的研究。 中药小蓟为菊科植物刺儿菜Cirsium Setosum(Willd.)MB.的干燥地上部分(《中国药典》(2000版)收载),具有凉血止血,祛淤消肿的功能。可用于治疗衄血,吐血,尿血,便血,崩漏下血,外伤出血,痈肿疮毒等,为历代中医临床的常用药之一。其化学成分包含黄酮类、生物碱类、有机酸类、皂苷类等,主要成分为黄酮类及有机酸类。 根据中医药理论,通过化学分离与药效学筛选比较,确定了小蓟炭饮片的主要有效部位即总黄酮部位。 采用分光光度法、高效液相色谱法检测小蓟中总黄酮和黄酮类化合物蒙花苷的含量,并进行了相应的方法学考察。以《中国药典》和《全国中药炮制规范》为主要参考,结合炮制历史沿革研究和现代文献研究,对小蓟饮片的炮制工艺进行优选。以蒙花苷及水分含量为指标,选择干燥温度、干燥时间2个因素,用L9(3)4正交设计表,采用方差分析法,确定具体的工艺技术参数,得小蓟饮片的最佳炮制工艺参数为:除去杂质,抢水洗净,80℃,干燥1小时。以总黄酮为指标,选择炒制温度、炒制时间2个因素,用L9(3)4正交设计表,采用方差分析法,确定具体的工艺技术参数。得炒小蓟的最佳炮制工艺参数为:260℃,炒制5分钟。为产业化、炮制方法的确定及申报小蓟饮片批准文号提供科学依据。 进行了小蓟及小蓟炭饮片的质量标准规范化初步研究,提出了小蓟及小蓟炭饮片的质量标准建议草案。 建立了小蓟的高效液相色谱指纹图谱,并通过指纹图谱相似度计算软件建立了小蓟指纹图谱的共有模式,对收集到的样品进行了相似度比较。并比较了小蓟饮片和小蓟炭饮片的差异。 上述研究结果为制定小蓟饮片炮制工艺及质量标准提供了科学的理论依据。

【Abstract】 The subject of Studies on Processing Technology and Quality Standard Research of herb of herba Cirsii was the subsidiary subject of a national "ten-five" major technology project-Standardization research of 50 kinds of traditional Chinese medical herb in pieces. The author studied systematically and deeply the processing history, contemporary documents, effective contents, standardized processing technology, quality standard and fingerprint of herb of herba Cirsii, which was often used in clinical research.The clinical practice has proved that herb of herba Cirsii has significant effect in cooling the blood and stop the bleeding and promoting the subsidence of swelling.The herba Cirsii are used in Chinese herbal medicine to treat constipation and improve visual acuity. Its chemical ingredients include flavonoids alkaloids organic acids. The main ingredients of it are flavonoids and organic acids.Based on the theory of traditional Chinese medicine, we demonstrated the effective part of herb of herba Cirsii and herb of herba Cirsii carbonisatus through the chemical separation and pharmacodynamical screening as flavonoids .The thesis determined the total contents of flavonoids in herb of herba Cirsii carbonisatus and linarinin herb of herba Cirsii carbonisatus by spectrophotometry and HPLC.With the study of the processing history and contemporary documents, indicated by contents of linarin and water, the optimal processing technology with water was identified with L9(3)4 orthogonal design, the factors of which were drying time and drying temperature used separately. The technology parameters were described as follows: pick out the impurity and clean quickly, then dry at 80℃ for 1 hour. Indicated by contents of total flavonoids, the optimal stirring technology was identified with L9(3)4 orthogonal design, the factors of which were stirring time and stirring temperature used separately. The technology parameters were described as follows: stir at 260℃ for 5 min.The thesis also carried out quality standard research of the herb of herba Cirsii and herb of herba Cirsii carbonisatus.Based on principles of fingerprint chromatography of Chinese herbal medicine, the optimal extraction method for herb of herba Cirsii and its corresponding HPLC-based fingerprint chromatography have been established through HPLC gradient elution. By means of similarity calculation and main ingredients analysis with fingerprint chromatography software, the common pattern of the herb of herba Cirsii s was established. The thesis also compared the similar degree of the herb of herba Cirsii from different distinct and the fingerprint chromatography of the herb of herba Cirsii and processed product of it.
