针刺对部分背根切断猫脊髓及备用背根节中CNTF表达的影响

【摘要】 目的:探讨CNTF、在正常猫、部分去背根猫及部分去背根猫针刺后备用背根节的表达情况,以了解CNTF与脊髓损伤修复及针刺促进脊髓损伤修复的关系。 方法:成年健康雄猫70只,随机分为七组,即正常对照组,手术非针刺3d组;手术非针刺7d组;手术非针刺14d组;手术针刺3d组;手术针刺7d组;手术针刺14d组。每组10只。手术组行单侧部分背根切除术(切除左侧L1~L5,L7~S2背根节,保留L6背根为备用根)。术后当天行针刺组L6脊神经外周支分布区域的穴位(足三里和悬钟、伏兔和三阴交)针刺,每天一次,每次30分钟,两组穴位交替进行。术后动物针刺及分别存活至3d、7d和14d时,各组取5只动物行4%多聚甲醛心内灌注固定。取单侧备用背根节进行-20℃恒冷箱切片,片厚20μm。分别用兔抗CNTF(1∶500)行免疫组织化学ABC法染色。观察CNTF在备用背根节及脊髓(L3,L6和背核)的分布及在术后不同时间的表达情况。进行组间多个样本均数的单因素方差分析。每组另取5只进行western blot检测在备用背根节及脊髓背角的CNTF蛋白量的变化。 结果 1.部分背根切断及针刺后,CNTF在备用背根节和脊髓的表达变化。部分背根切断后的表达变化:部分背根切断后3d,非针刺组脊髓(L3,L6和背核)CNTF阳性细胞和背根节的CNTF总、中小阳性神经元数较正常组明显减少(P<0.05)。7d、14d背根节的CNTF总、中小阳性神经元数与正常组比较无差异(P>0.05),而非针刺组大阳性神经元数在各个时间段与正常组比较均无差异(P>0.05)。L6

【Abstract】 Objective: To evaluate the plastic changes of CNTF in the left sixth lumbar dorsalroot ganglion (L6 DRG), spinal cord in representative spinal segments L3, and L6 and in the nucleus dorsalis (ND) at L3 segments after acupuncture in cats subjected to removal of L1-L5 and L7-S2 DRGs.Method: Senvety male adult cats were divided into seven groups (n=10 in eachgroup), and every five cats were used for immunohistochemistry and western blot respectively in all groups. Group I were normal control animals. Groups II, III and IV cats were subjected to surgical removal of the left L1-L5 and L7-S2 DRG, but sparing the left L6 dorsal root ganglioa GroupsV, VI and VII cats were first subjected to the aforesaid ganglionectomy, then followed immediately with electrical stimulation on the operated side at acupuncture points (xuewei 穴位) zusanli-xuanzhong (足三里-悬钟), and futu-sanyinjiao (伏兔-三阴交), known to lie in L6 dermatome. They were allowed to survive for 3(Groups II and V cats), 7(Groups III and VI cats) or 14 (Groups IV and VII cats) days after operation. Surgery was performed under deep anesthesia achieved with an intraperitoneal injection of 3.5% pentobarbital sodium (1.3 ml/kg body weight). All the DRG together with a small segment of the respective dorsal roots measuring 3 mm long from L1 through S2, sparing the L6, were removed through the intervertebral foramina.
