北柴胡地上部分生物活性成分的研究

【摘要】 伞形科柴胡属(Bupleurum L.)植物北柴胡(Bupleurum chinense DC.)作为药用植物已有悠久的历史,是以根入药为主的常用传统中药,茎叶多弃之不用,造成资源利用不足。为了开发药源,我们对其地上部分的化学成分进行研究,为北柴胡药用部位的选取,提供化学上的依据。通过溶剂提取及各种色谱方法(大孔吸附树脂、硅胶、凝胶、中性氧化铝等柱色谱),从北柴胡地上部分的热水提取物中分离得到11个化合物,通过波谱分析、理化常数测定并结合文献,鉴定了其中10个化合物,分别为柴胡素(bupleurin Ch-1),异鼠李素(isorhamnetin Ch-2),异鼠李素-7-O-α-L-鼠李糖苷(isorhamnetin-7-O-α-L-rhamnopyranoside Ch-3),山柰酚(kaempferol Ch-4),山柰酚-3-O-α-L-阿拉伯糖苷(kaempferol-3-O-α-L-arabinofuranoside Ch-5),山柰酚-7-O-α-L-鼠李糖苷(kaempferol-7-O-α-L-rhamnopyranoside Ch-6),山柰酚-3,7-二-O-α-L-鼠李糖苷(kaempferol-3,7-di-O-α-L-rhamnopyranoside Ch-7),芦丁(rutin Ch-8),2,5-二甲基-7-羟基色原酮(7-hydroxy-2,5-dimethyl-chromnone Ch-9),β-谷甾醇(β-sitosterolCh-10)。其中化合物Ch-1为一新化合物;化合物Ch-5和Ch-9均为柴胡属首次分离得到。 利用HPLC同时测定了北柴胡地上部分中的两种活性成分山柰酚和山柰酚-7-O-α-L-鼠李糖苷的含量,考察了稳定性、重现性、精密度等条件,为药材质量控制提供了可能。 生物活性实验显示北柴胡地上部分中的单体化合物均能显著升高培养液中NO2-的浓度,且Ch-2(1μg/ml)、Ch-3(1μg/ml)、Ch-7(0.1μg/ml、1μg/ml、10μg/ml)能加强LPS刺激巨噬细胞产生NO。其中除Ch-6的高剂量(10μg/ml)外均不影响细胞成活率。
【Abstract】 In this paper, in order to obtain the chemical constituents from Bupleurum chinense DC. The boiling water extract of Bupleurum chinense DC. was studied. Eleven compounds were isolated and purified by various chromatographic methods, such as macroporous resin column chromatography, silica gel column chromatography, sephadex LH-20 column chromatography and so on, and their structures were established by chromatographies and spectral analysis. They were elucidated as following: bupleurin(Ch-l), isorhamnetin (Ch-2), isorhamnetin-7-O-α-L-rhamnopyra-noside (Ch-3), kaempferol (Ch-4), kaempferol-3-O-α-L-arabinofuranoside (Ch-5), kaempferol-7-O-α-L-rhamnopyranoside (Ch-6),kaempferol-3,7-di-O-α-L-rhamnopyran-oside (Ch-7), rutin (Ch-8), 7-hydroxy-2,5-dimethyl-chromnone (Ch-9), β-sitosterol (Ch-10). Among them, compound Ch-1 is a new compound; compound Ch-5 and compound Ch-9 were isolated from the genus for the first time.The bioactivited contents of kaempferol and kaempferol-7-O-α-L-rhamnoside were determined by HPLC at the same time. And it leads to the possibility of the quality control.Bioactivity experiment indicated that the compounds from Bupleurum chinense DC. significantly increased the concentration of NO2- in the medium. And Ch-2(1 ug/ml), Ch-3 (1 wg/ml), Ch-7 (1 wg/ml) can enhance the induction of NO in macrophages by LPS. All the extracts can not influence the cell survival except for Ch-6 (10 wg/ml).
