文拉法辛等药物的手性分离与药物动力学研究

【摘要】 手性是生物体系的一个基本特征。手性药物对映体通过手性识别机制与体内内源性大分子物质结合,产生一系列生理生化反应,由于立体结合位点的不同而产生不同的药动学特征和药理学作用。手性药物的分离测定,对于研究药物的体内药动学过程,研究其药理和毒理作用机制以及质量控制都具有重要的意义。本文采用高效液相色谱手性固定相法分离了文拉法辛、布洛芬、曲美布汀、福多司坦四种手性药物,并研究了文拉法辛在动物体内的立体选择性药动学过程。 1.文拉法辛、布洛芬、曲美布汀的对映体分离方法研究 文拉法辛、布洛芬、曲美布汀在临床上均以消旋体的形式给药。为研究其手性分离方法,采用高效液相色谱法,使用α1-酸性糖蛋白手性固定相,通过考察有机改性剂的种类、浓度、流动相的pH值、缓冲溶液的浓度、流速等因素对对映体分离的影响,优化了色谱条件。文拉法辛等三种手性药物均在α1-酸性糖蛋白固定相上得到了完全分离。 2.福多司坦的右旋异构体检查方法研究 临床上使用左旋福多司坦作为祛痰剂,右旋体为杂质需控制其含量。福多司坦由于其特殊的分子结构,不能使用α1-酸性糖蛋白手性固定相进行对映体分离。本文用Chirobiotic T手性固定相分离了福多司坦对映体,并建立了福多司坦右旋体异构体的检查方法,该方法灵敏度高,重现性好,可用于原料药和制剂中右旋福多司坦的限量检查。 3.抗抑郁药文拉法辛在大鼠体内的药动学研究 建立一种简单、灵敏的高效液相色谱方法测定大鼠血浆中的文拉法辛,并对其药动学进行了研究。以替硝唑为内标,血浆样品碱化后,经液—液提取在C18反相色谱柱上进行分离,检测波长为230nm。文拉法辛的线性范围为0.156~20μg/mL,最低定量限为0.156μg/mL,日内精密度RSD为2.8%~5.2%,日间精密度RSD为4.2%~5.6%,准确度RE值为-2.7%~4.0%,该方法适用于大鼠体内药动学研究。大鼠灌胃给予单剂量文拉法辛120mg/kg后,测定血浆中文拉法辛浓度,得到的药动学参数表明,该药吸收迅速,平均达峰时为0.73h,平均峰浓度为12.3μg/mL,血药浓度—时间曲线下面积AUC0-t采用线性梯形法估算,平均值为44.3μg·h/mL,大鼠灌胃给药的药时过程符合二室模型。 4.大鼠血浆、脑组织中文拉法辛对映体的测定 采用非手性、手性色谱系统相结合的方法,建立了测定大鼠血浆和脑组织中文拉法辛对
【Abstract】 Chirality is a fundamental phenomenon that plays an important role in biological processes.A wide range of biological and physical functions are generated through molecular recognition because endogenous substances within biological systems interact with different enantiomers in decisively different ways. As a result of such chiral recognition,drug enantiomers may differ in their pharmacokinetic characteristics and pharmacological effect. The availability of chiral analytical separation methods are important in studying and understanding the pharmacokinetic processes ,the pharmacological and/or toxicological effects and quality control of many drugs.In this paper, HPLC methods using chiral stationary phases(CSP) have been developed for separation of four drugs : venlafaxine , ibuprofen, trimebutine and fudosteine.The enantioselective pharmacokinetics of venlafaxine in rat plasma have been studied.1.Studies on the separation methods of enantiomers for venlafaxine , ibuprofen and trimebutineClinically,venlafaxine , ibuprofen and trimebutine are all administered as racemates. Influences of the pH value of the mobile phase,the organic modifiers,the buffer concentration,the flow rate and the column temperature on the enantiomeric separation of the three drugs by HPLC using α1-acid glycoprotein(α1-AGP) CSP were investigated and the enantiomeric separation were optimized .Venlafaxine , ibuprofen and trimebutine enantiomers can be baseline separated on the ai-AGP column. 2.Studies on chiral HPLC determination of dextro enantiomer of FudosteineLevo-fudosteine is a mucoactive agent but its dextro enantiomer has no activity,so dextro-fudosteine needs quality control as impurity.Fudosteine enantiomers cannot be separated on an ai-AGP CSP because its chemical stracture.A HPLC method using chirobiotic T CSP has been developed for chiral separation and determination of dextro enantiomer of fudosteine.The study provides a simple and reproducible method for the quality evaluation and control of dextro-fudosteine in material and formulation.
