脉通颗粒的制备工艺及质量标准研究

【摘要】 目的:制定脉通颗粒的制备工艺和其质量控制标准。 方法:通过L9(34)正交试验优选脉通颗粒的最佳提取工艺、最佳精制工艺,并对结果进行方差分析,最终确定试验参数。水提工艺研究中就影响提取物质量的重要因素:提取次数(A)、煎煮时间(B)和加水量(C)进行了考察,以丹酚酸B和总浸出物为指标进行工艺优选。精制工艺中以丹酚酸B为指标,对影响的主要因素:相对密度(A)、醇沉浓度(B)、醇沉温度(C)进行了正交试验。此外还对该颗粒剂进行了成型性研究和筛选,以成型性、堆密度、休止角、溶化性、吸湿性和临界相对湿度为指标,考察糊精、淀粉和乳糖三种辅料在制粒中的优劣性,并用综合评分法筛选出制备颗粒剂的最优辅料组成。考察了矫味剂甜蜜素、甜菊糖和蛋白糖的用量和口感,并最终确定矫味剂的种类和用量。采用己确定的工艺条件,试制三批中试样品,并对其进行检验,制定出脉通颗粒的质量标准,从而为大生产提供合理可行的技术参数。通过薄层定性鉴别脉通颗粒中的黄芪、丹参、赤芍药材;同时采用薄层扫描法对君药黄芪所含的活性成分黄芪甲苷进行含量测定,采用高效液相色谱法对臣药丹参及佐药赤芍所含的的有效成分丹酚酸B和芍药苷进行含量测定。 结果:水提工艺中三因素均对质量有影响,其影响大小以丹分酸B含量为考察指标,各因素对其含量影响的大小顺序为B>A>C;以总浸出物为考察指标,各因素对其影响的大小顺序为A>B>C,经方差分析后,最终确定脉通颗粒的优选工艺为A281C2,即加10倍量的水,煎煮2次,每次1小时。精制工艺各因素对丹酚酸B的影响大小顺序为A>B>C,最终确定其醇沉工艺为A181C2,即浓缩至相对密度为1.05~1.10,待药液放置温度为50℃时,加入一定量的乙醇,使其药液含醇量为60%即可。成型性试验结果确定脉通颗粒的选用辅料以乳糖为主,再加入少量的甜菊糖作为矫味剂。薄层鉴别结果脉通颗粒中黄芪、丹参、赤芍均斑点清晰,阴性无干扰。含量测定结果黄芪甲苷进样量在0.504μg~2.52μg范围内与峰面积线性关系良好(r=0.999),平均加样回收率为99.18%(RSD=1.84),并制定本品含黄芪以黄芪甲苷(C41H68O14)计每克不少于0.3mg。丹酚酸B进样量在0.16μg~0.80μg范围内与峰面积线性关系良好(r=0.9999),平均加样回收率为100.43%(RSD=1.49),并制定本品每克含丹参以丹酚酸B(C36H30O6)计,不得少于1.1mg。芍药苷进样量在0.132μg~0.66μg范围内与峰面积线性关系良好(r=0.9999),平均加样回收率为98.87%(RSD=1.74),并制定本品含赤
【Abstract】 Objective: To establish the preparation craft and quality control standard of Maitong Granule.Methods: L9(34) Orthogonal design was used to optimize the best extraction craft and the best refined craft of Maitong Granule, and carry on analysis of variance to the result, confirm the experimental parameter finally. In water extraction craft research, the most important influence factor for the extraction quality are extraction times(A), decoction times(B)and adding the amount of water(C), and carry on the investigation with the Salvianolic acid B and total immersion for target, carrying on the craft optimization with the Salvianolic acid B for target in refined craft, making orthogonal experiment for the most factor:relative density(A) , ethanol deposit chroma(B)and ethanol deposit temperature(C). shaping research and screen to this guanule in addition, with shaping, pile density, angle of repose,dissolving, hygroscopicity and borderline relative humidity as the target, invegate the advantages and disadvantages of dextrin, starch, lactose in making guanule, and select the optimum adjuvants which prepare guanule to make up with the comprehensive point system. Investigate and rectify flavor pharmaceutical sodium cyclamate, stevioside and aspartame consumption and mouth feel of candy, and confirmFinally that rectf ies kind of pharmaceutical of flavor and consumption. Regard craft parameter confirmed as the standard, trial produce three batches of pilot scale, and examine it, make the quality stadard of Maitong guanule, offer reasonable feasible technical parameter for great prduction Radix astragali, Radix Salviae miltiorrhizae , Radix paeoniaerubra were identified for qualitative identification by TLC, and the content of Astragaloside in monarch Radix astragali was determined by TLCS, the content of Salvianolic acid B in official drug Radix Salviae miltiorrhizae and Paeoniflorin inadjuvant drug Radix paeoniae rubra were identified by HPLC.
