水牛胚胎体外培养影响因素的研究

【摘要】 本研究主要探讨氨基酸和维生素对水牛体外受精(IVF)和孤雌激活(PA)胚胎体外发育的影响,同时对不同品牌石蜡油的水牛孤雌激活胚胎体外培养效果进行了比较。 在SOF液中添加1.5%的MEM非必需氨基酸(NEAA)显著提高PA(79.8%vs67.7%)和IVF胚胎(65.7%vs51.9%)的分裂率(P<0.05),但添加3.0%的BME必需氨基酸(EAA)则显著降低IVF胚胎的分裂率(42.2%vs52.2%),虽然添加1.0%的EAA显著提高PA胚胎的囊胚孵化率(77.3%vs43.3%,P<0.05)。添加1.0%的MEM维生素显著提高IVF胚胎的分裂率(65.6%vs49.4%,P<0.05),但显著降低PA胚胎的囊胚发育率(13.9%vs26.7%,P<0.05)和囊胚孵化率(28.0%vs50.0%,P<0.05)。通过比较了几种不同来源石蜡油(协和油、Ameresco、Sigma胚胎培养油和Sigma过滤油)的水牛PA胚胎体外培养效果发现,Ameresco油的7天囊胚发育率(22.9%)和总囊胚率(28.8%)均显著低于Sigma过滤油(39.9%和42.8%)和Sigma胚胎培养油(34.4%和40.2%)(P<0.05),虽然各组间分裂率和囊胚孵化率无显著差异。 以上结果表明:(1)非必需氨基酸对水牛PA和IVF胚胎的体外发育都具有一定的促进作用;(2)低浓度的必需氨基酸对水牛胚胎的发育具有一定促进作用,但高浓度则有抑制作用;(3)维生素对水牛PA胚胎发育有抑制作用,但对IVF胚胎则有促进作用;(4)石蜡油对胚胎培养效果有一定影响,Sigma油可满足胚胎培养的需要。
【Abstract】 Effects of amino acids and vitamins on the in vitro development of buffalo embryos derived from parthenogenetic activation(PA) or in vitro fertilization( IVF) were investigated, and the effects of the mineral oil sources on the in vitro development of buffalo embryos derived from parthenogenetic activation(PA) were also examined in this study.Addition of 1.5% MEM nonessential amino acid solution (NEAA) to SOF medium resulted in a significant increase in the cleavage rate of either PA (79.8% vs 67.7%, P<0.05) or IVF embryos (65.7% vs 51.9%, .P<0.05). However, supplementation with 3.0% BME essential amino acid solution (EAA) decreased the cleavage rate of IVF embryos (42.2% vs 52.2%, P<0.05), although the hatched rate of PA blastocysts was increased by supplementation with 1.0% EAA (77.3% vs 43.3%, P<0.05). Addition of 1.0% MEM vitamin solution (Vit) to SOF medium increased the cleavage rate of IVF embryos (65.6% vs 49.4%, P<0.05)> but decreased the proportion of PA embryos developed to blastocysts (13.9% vs 26.7%, P<0.05) and hatched blastocysts (28.0% vs 50.0%, P<0.05). Significantly more PA embryos cultured with Sigma filterd oil or Sigma embryo tested oil developed to blastocysts on Day 7 (39.9% or 34.4%) and by Day 9 (42.8% or 40.2%) in comparison with Ameresco mineral oil (22.9% and 28.8%, P<0.05).In conclusions, (1) nonessential animo acids can stimulate the development of buffalo embryos derived from either PA or IVF; (2) Low concentration of essential animo acids is beneficial to the development of buffalo embryos but high concentration of EAA is detrimental to the development of buffalo embryos; (3) Vitamins are beneficial to buffalo IVF embryos but detrimental to PA embryos; (4) The in vitro development of buffalo embryos was influenced by the quality of mineral oil and the mineral oil from Sigma is suitable to embryo in vitro culture.
