肝疏泄太过深层机制探索-PMS肝气逆证猕猴模型中枢关键受体基因差异表达研究

【摘要】 目的:以肝疏泄太过典型病证经前期综合征(Premenstrual syndrome,PMS)肝气逆证为切入点,探寻与该病证发病密切相关的中枢内单胺类神经递质与甾体激素受体基因,探索研究肝疏泄太过深层机制的途径,在基因表达水平检验“肝主疏泄与调节机体单胺类神经递质、性激素及其调节激素水平有关”科学假说。方法:利用已建立的正常猕猴与PMS肝气逆证模型猕猴脑组织正向、反向差减cDNA文库,制备表达谱基因芯片寻找差异表达基因。结果:中枢单胺类神经递质受体5HT1A受体、5HT2A受体在PMS猕猴模型边缘系统中mRNA表达均升高;GABAA受体在猕猴模型边缘系统中mRNA表达降低。中枢甾体激素受体ERα、ERβ在PMS猕猴模型边缘系统mRNA表达降低;PGR在PMS猕猴模型边缘系统mRNA表达升高。结论:支持“肝主疏泄与单胺类神经递质、调节性激素水平有关”科学假说,上述六个受体基因与PMS肝气逆证中枢发病机制关系密切。

【Abstract】 Objective: Premenstrual syndrome (PMS) liver-qi sthenia symptom is the type disease of Liver-qi Sthenic Dispersedness, which is the key point to study and explore nerve center monoaminergic neurotransmitters and hormone receptor genes. With its close relateness to the Underlying Mechanism of Liver-qi Sthenic Dispersedness,to explore the genes and the approach how it acts,furthermore,detect“liver-qi sthenic symptom complex of Traditional Chinese Medicine related deep mechanism”and“liver-qi sthenic symptom complex of Traditional Chinese Medicine related monoaminergic neurotransmitters and hormone”scientific hypothsis in gene level. Methods:Utilize the used subtractive cDNA library,make Macroarray gene chip to search different expression of genes.Results:nerve center monoaminergic neurotransmitters 5HT1AR、5HT2AR mRNA increase and GABARA mRNA decrease in the margin systen of PMS macaque model. hormone ERα,ERβmRNA decrease and PGR mRNA increase in the margin system of PMS macaque model. Conclusion:sustain scientific hypothsis,six candidacy genes are greatly related to PMS liver-qi sthenic symptom nerve center mechanism.
