南京椴种子生物学特性与休眠机理初探

【摘要】 本文主要以分布于安徽皇藏峪和南京林业大学校园内的南京椴为研究对象,对其种子生物学进行观察和分析,对种子的休眠机理进行多方面的研究,结果表明:1、对南京椴开展种子生物学特性分析和测定表明,该种子实为果实,中果皮高度木质化,种皮密被蜡质,果皮、种皮坚硬,并对种子吸水有一定阻碍作用;片林种子饱满度较高,孤立木种子饱满度极低,同时不同产地种子大小和重量也存在差异。2、各种方式层积处理带壳种子效果均不佳, L 9 ( 3 4 )正交试验获得去壳种子打破休眠最佳处理组合,同时也是目前最优处理方法为:70℃热水浸泡种子20min,1000mg·L-1GA3溶液浸泡6h,而后5℃低温层积60d,种子发芽率可达14%。3、试管培养表明,胚乳中存在抑制种子萌发的关键性因素,剥除胚乳或赤霉素处理可以消除胚乳对种胚萌发的抑制,种胚中不存在抑制种子萌发的决定性因素。4、抑制物提取测定表明,果壳、种皮、胚乳皆含有抑制物,果壳中抑制物分布于乙醚相和甲醇相中,种子部分抑制物存在于甲醇相中,种子甲醇相所含抑制物抑制活性最强。5、通过对种子激素含量、营养物质含量、氧化途径关键酶活性测定,结果表明种子在破除休眠过程中,GA3含量和GA3/ABA快速升高,ZR/ABA缓慢升高;可溶性糖含量升高,可溶性蛋白含量升高再降低;PPP途径和TCA循环均呈逐渐活化的趋势。激素调控学说、代谢活性学说、氧化途径学说皆适于解释南京椴种子休眠机理,但都难以完美。

【Abstract】 This article mainly take the Nanjing Linden distributed in Emperor hided valley of Anhui Province and Nanjing Forestry University campus as the research object.The Seed biological characteristics were observed and analysed,the seed dormancy mechanism were widespread researched.Results showed,1、The seed biological characteristics showed that the seed is fruit actually,the mesocarp turned linnification,the seed coat is densely covered by the wax,the fruit shell and the seed coat are very hard,and have the certain hindrance function to seed absoring water.The seed plumpness of greenbelt was higher,the seed plumpness of single tree was extremely low. Meanwhile,the size and the weight of seed from different place of production also have the difference.2、Each kind of stratification can not broke the dormancy of the seed with fruit shell. Under the orthogonal L9(34)experiment,obtained the optimum approach of hucked seed to break the dormancy is 70℃hot water soaked seeds 20min,1 000mg·L-1GA3 solution soaked 6h,then 5℃low temperatures stratificated 60d,the germination rate can reach to 14%, which was the best method of break the Nanj ing Linden seed dormancy at present.3、The germination in vitro indicated that,the key factor of inhibiting seed germination is in the endosperm.Removed the endosperm or treated the kernel with gibberelin solution can eliminate the suppression of endosperm.The embryo do not have the determining factors which restrained seed germination.4、The growth inhibitor of Nanjing Linden seed extracted and bioassied showed,growth inhibitor were all contained in the fruit shell,seed coat,and endosperm.The growth inhibitor of fruit shell distributed in the phases of Ether and the phases of Methanol,while the growth inhibitor of seed distributed in the phases of Methanol which inhibit activity is the strongest.
