中药DGH胶囊对自发性糖尿病大鼠玻璃体血管生长因子及视网膜形态学损害影响的实验研究

【摘要】 目的：观察中药DGH胶囊对6个月病程的自发性糖尿病大鼠玻璃体血管生长因子及视网膜微血管损害的影响，探讨中医药治疗糖尿病视网膜病变的病理机制。 方法：采用6个月病程的自发性糖尿病大鼠模型，随机分为6组：阴性对照组、中药DGH胶囊低、中、高剂量组、递法明组及导升明组，另设有正常对照组。连续灌胃3个月，并于灌胃3月末处死大鼠，通过酶联免疫及免疫组化等方法，观察中药DGH胶囊对DM大鼠玻璃体生长因子及视网膜微血管损害的影响。 结果：(1)中药DGH胶囊有改善DM大鼠生存状态的作用。(2)中药DGH胶囊可以降低糖尿病大鼠玻璃体胰岛素样生长因子-1的水平，从而对新生血管的形成起到抑制作用。(3)中药DGH胶囊可以抑制视网膜血管基底膜Ⅳ型胶原的增生，且作用优于导升明和递法明组，从而对糖尿病视网膜微血管损害起到抑制作用。(4)中药DGH胶囊对DM大鼠血糖有一定降低作用。 结论：中药DGH胶囊能抑制视网膜血管基底膜Ⅳ型胶原的增生，降低糖尿病大鼠玻璃体胰岛素样生长因子-1的水平，对糖尿病视网膜微血管损害有一定的保护作用，对新生血管形成有抑制作用。更多还原

【Abstract】 Objective:to study the effects of DGH on the growth facter in vitreous and retinal microvascular impairment of the experimentally GK rat,a Spontaneous Model of Non-Insulin-Dependent Diabetes, and to explore the pathology mechanism of how the Chinese Medicine treating DR.Methods:After observed 6 months ,GK rats were randomly divided into 6 groups according to the weight and the level of blood glucose: model group ,treated groups with high、middle and low dosage of DGH ,positive control group of Difral and Doxium ,also there are normal group. After given drugs or distilled water for 3 months, the rats were put to death , eyeballs of the rats were taken out. Their vitreous and retinas were studied by means of ELISA and immunohistochemistry respectively.Results:(1) DGH has the effect of improving the existing behavior of GK rats. (2) DGH can reduce the level of insulin-like growth factor-1 of GK rats in vitreous. (3) DGH can inhibit the proliferation of type IV collagen of retinal capillory basement membrane of GKrats. (4)DGH can decrease the blood glucose of GK rats in some degree.Conclusion: DGH capsule can inhibit the proliferation of type IV collagen of retinal capillory basement membrane of GK rats and decrease the level of insulin-like growth factor-1 of GK rats in vitreous , so it is a good capillary protective agent to the GK rats’retinal impairment.更多还原
