化瘀行水验方拮抗肾小球硬化进展的实验研究

【摘要】 目的：探讨化瘀行水验方拮抗5／6肾切除大鼠肾小球硬化的作用。 方法：随机将动物分为空白对照组，5／6肾切除大鼠模型对照组和化瘀行水验方组。分别于手术后4周、8周检测各组大鼠血红蛋白、血肌酐、血尿素氮和24h尿蛋白含量；Masson和免疫组化染色结合图像分析系统半定量观察肾小球内转化生长因子β₁（TGF-β₁）、纤维连接蛋白（FN）、层粘连蛋白（LNα-1）、Ⅲ型胶原（Coll Ⅲ）、Ⅳ型胶原（Coll Ⅳ）及胶原纤维的积分光密度（IOD）。 结果：模型组较空白对照组血肌酐、血浆尿素氮和24h尿蛋白明显增加（p＜0.001），而血红蛋白明显减少（p＜0.001），这种改变随着时间延长而逐渐加重。中药组较模型组治疗4周、8周后，能显著地减少血肌酐、血尿素氮含量（p＜0.05），显著减少24h尿蛋白量（p＜0.05～0.01）。但不能明显改善血红蛋白的减少（p＞0.05）。化瘀行水验方治疗4周及8周后，肾小球内TGF-β₁、FN、LN、Coll Ⅲ、Coll Ⅳ和胶原纤维的病理性聚集显著减少（P＜0.05～0.01）。 结论：化瘀行水验方治疗后，其减少5／6肾切除大鼠24h尿蛋白量和拮抗肾小球硬化的作用，至少部分参与了保护残存肾功能的作用。更多还原

【Abstract】 OBJECTIVE: To investigate the effects of preventing progress of glomerulosclerosis by the Huayu Xingshui recipe in rat with 5/6 nephrectomy.METHODS: The rats were randomly assigned to Huayu Xingshui recipe treatment group, control group and normal group. Hemoglobin, serum creatinine （Scr）, blood urea nitrogen （BUN） and 24h urinary protein were me sured at 4th and 8th week after the nephrectomy. Renal pathology changes were evaluated at the same time. The renal tissues were stained by immunohistochemical and Masson methods. Then integra optical densit（IOD） of TGF-β₁, FN, LNα -1, Coll III, Coll IV and Collagen fibrin of the glomerular were semi-quantified by image analysis system.RESULTS: In comparison with the control group, Scr , BUN and the 24h urinary protein were decreased in Huayu Xingshui recipe group（P<0.05）. But the hemoglobin can not be prevented decreasing significantly （P>0.05）. The immunohistochemical result showed that TGF- β₁, FN, LN α -1, Coll III,Coll IV and Collagen fibrin of the glomerular were significantly decreasedin Huayu Xingshui recipe group （P<0.05—0.01） .CONCLUSION: After the treatment of Huayu Xingshui recipe, the renal function were protected leastways partly by the participant of decreasing 24h urinary protein and preventing glomerulosclerosis.更多还原
