川陕两地肾阳虚证的免疫功能基因初探

【摘要】 背景 近年来,由于医学模式的改变,尤其是人类结构基因组计划的顺利完成,以及功能基因组学的快速发展,为中医证候的研究带来了新的机遇,如何进行证的现代化研究是目前中西医结合研究的热点,但由于证的复杂性,故以高通量基因芯片技术为桥梁,可望整体地、系统地研究证的功能基因组,本文以两批肾阳虚证初步探讨其免疫功能基因组,虽然这还仅仅是人类功能基因组研究的冰山一角,但是对于解决目前证的研究中面临的困惑具有一定的积极的意义。 目的 (?)研究肾阳虚证的差异基因表达谱; (?)寻找与肾阳虚证相关的免疫功能基因组。 方法 (?)理论:在“证候为特定症状组合的一群功能基因的异常表达”的假说基础上,我们以NCBI及GO数据库为基础、中医辨证论治为指导思想、现代医学模式为参照、按病证结合的方法,研究肾阳虚骨关节炎患者的基因表达谱,着重从现代免疫学的角度分析,寻找相关的免疫功能基因组,以研究免疫基因在肾阳虚证中的变化情况。 (?)实验:以40个量的肾阳虚量表,精选川陕两地两批共24人(各以8例骨关节炎肾阳虚证患者,8人为正常对照组),22000点的六张基因芯片(单体样本4份,合体样本2份)为研究对象,参照西医病名及理化手段、转录组学的基因表达枝术、数学上的生物统计、生物信息学的信息挖掘等多学科结合,研究肾阳虚证的差异基因表达谱,寻找相关的免疫功能基因组。 结果 (?)系统地分析了16位肾阳虚患者的证候评分表以及正常对照组的肾阳虚证候表。 (?)经手工查找获得66条在六张芯片中重复表达且达到差异倍数基因;经进一步信息挖掘,有61条可在网络数据库里被识别,即有61条已知功能的基因,占所有重复表达差异基因的92.42%。发现有22条基因与免疫相关,占已知功能基因的36.07%,说明肾阳虚证与免疫基因表达有着密切的关系。 (?)在网络基因数据库里分析六张基因芯片的生物学功能和涉及的相关通

【Abstract】 BackgroundAfter the Human Genome Project to be completed, the arrival of post genome era provides a new opportunity to study the Zheng of Traditional Chinese Medicine (TCM). At present, how to explore "Zheng" are the important focuses of combination of TCM with Western medicine research. Because of the complexity of Zheng, we used the method which link disease with Zheng. Microarray technology is a way of building bridges between disease and Zheng. It has positive meanings to clarify many problems in the current Zheng study.Objective① To study the differential expression genes of deficiency of kidney Yang ② To research differential expression immune gene of deficiency of kidney YangMethods① Based NCBI database and GO database, taked the TCM Bianzhenglunzhi as the guiding ideology, refer to the modern medical model, combined the disease with Zheng (disease called by Western medicine and Zheng on TCM), we studied the microarray of patients who have osteoarthritis and deficiency of kidney Yang. We focused on the view of modem immunology, searched for relevant immunogenes, and the changes of immunogene in the patients who have deficiency of kidney Yang.② We studied the topic on many subjects such as TCM、 western medicine、 molecular biology、 biodemography、 bioinformatics. We picked the samples (16 patients with osteoarthritis and the deficiency of kidney Yang、 8 norm person as the control group)from 2 areas, made 6 microarrays (4 single samples and 2 compond samples), and studied the differential gene microarray of the patients with deficiency of kidney Yang, search for relant immunogene.Results① We systematically analyzed 16 patients’ Zheng scores who have deferency of kidney Yang and the control group’s Zheng scores.② We got 66 differential genes in six microarrays. Furthermore, after we put the 66 genes into network database, 61 genes can be identified in the network database. Among the 61genes, 22 genes are related to immune, which can tell us the close relationship between the deficiency of kidney Yang and immunogene expression.③ We analyse biological functions and pathways of the genes in 6 microarrays by using the network database.④ We established the genetic basis of the combination of disease and Zheng,
