绞股蓝多糖的分离纯化及其药理活性研究

【摘要】 绞股蓝Gynostemma pentaphyllum(Thunb.)Makino(GP)为葫芦科绞股蓝属多年生草质藤本植物,又称七叶参、七叶胆、甘茶蔓。全世界有16种和3变种,我国有15种和3变种,主要分布于长江以南各省区,其中以云南种类最多。关于绞股蓝的研究,上世纪九十年代文献报道较多,主要集中在绞股蓝皂苷成分上,较少涉及绞股蓝多糖(Gynostemma pentaphyllum polysaccharides,GPS)的研究,而且涉及绞股蓝多糖的也是绞股蓝粗多糖的研究,未对绞股蓝粗多糖进行分离纯化。根据文献报道,绞股蓝粗多糖的主要药理作用有抗肿瘤和增强免疫力两个作用,另外还有抗氧化等作用。本论文对绞股蓝粗多糖进行了分离纯化,得到了两种精制多糖组分,并对其结构和药理活性进行了初步研究。论文主要内容如下: 1、绞股蓝粗多糖提取方法的优化。绞股蓝药材粉碎后,先用乙醇回流除去单糖、寡糖等小分子物质,然后采用正交实验设计考察了料液比(A)、提取时间(B)、提取温度(C)和提取次数(D)对绞股蓝粗多糖提取率的影响,并结合实际综合考虑提取效率,多糖的性质等,最终确定最佳提取工艺A2B1C3D2,即:10倍量水、100℃、0.5小时提取3次。 2、绞股蓝多糖的分离纯化。绞股蓝粗多糖经氯化钙脱果胶,双氧水脱色得到纯化多糖。纯化多糖经DEAE-Sephrose分离,氯化钠溶液洗脱,紫外210nm结合苯酚硫酸法检测组分,得到GPS-2、GPS-3、GPS-4三个多糖组分。GPS-2、GPS-3、GPS-4各多糖组分再分别经Superdex G-200柱分离,0.15mol/L氯化钠溶液洗脱,紫外210nm检测,收集各组分,冷冻干燥,再经Sephadex G-25脱盐,最后冷冻干燥得绞股蓝精制多糖GPS-2、GPS-3、GPS-4。
【Abstract】 Gynostemma pentaphyllutn (Thunb.) Makino, a member of Cucurbitaceae family, is a kind of herb. There are 16 species and 3 varieties of Gynostemma pentaphyllum in the world, and 15 species and 3 varieties in China. They are distributed over large areas in the south provinces of China, especially in Yunnan province. In the 90s of last century, there were many reports about Gynostemma pentaphyllum, but they focused mainly on Gynostemma pentaphyllum saponins and only a few about Gynostemma pentaphyllum polysaccharides. Raw polysaccharidebs but not homogeneous polysaccharides was used in the previously research of Gynostemma pentaphyllum polysaccharides .According to the reports, the main pharmacological action of raw Gynostemma pentaphyllum polysaccharides included the action of anticancer and the action of enhancing immunity. In this papers, we separated and purified the raw Gynostemma pentaphyllum polysaccharides and obtained two homogeneous polysaccharides. We studied the structure and pharmacological action preliminarily. The main contents of the papers are as follows:1. The optimization of extraction method of raw Gynostemma pentaphyllum polysaccharides. First, monosaccharides, oligosaccharides and other micromolecule matters were removed by ethanol, then orthogonal design method was used to optimize the extracting conditions. Considering the extraction efficiency and quality of polysaccharides, the final optimization extracting method was A2B1C3D2.2.
