RAPD和ISSR标记对果蔗种质遗传多样性的研究

【摘要】 应用RAPD与ISSR技术对39份不同地方果蔗品种和外引黑皮果蔗Badila进行了种质遗传多样性分析。分别从100条RAPD引物和100条ISSR引物中筛选出适合果蔗种质分析的23条RAPD引物和28条ISSR引物用于RAPD和ISSR分析。23条RAPD引物共扩增出250条带,多态性条带数为175,多态性条带比率为70%;28条ISSR引物共扩增出301条带,多态性条带数为232,多态性条带比率为77.1%。基于扩增条带数据库建立了各自的Jaccard遗传相似系数矩阵,构建了趋势相似但不完全相同的分子树状图,将40份果蔗种质分为四组:第一类为已鉴定为杂交种的白鳝、歪干担、肚度、温岭果蔗以及人工杂交选育的果蔗品种474;第二类为外引黑皮果蔗Badila和丰城紫皮果蔗;第三类为单独的广东黄皮果蔗一类;第四类为32份地方果蔗品种,包括福建、江西、浙江、广西、云南等省的不同地方果蔗品种。两种分子标记的分析结果呈极显著相关(r=0.9471)。结果表明:RAPD与ISSR标记适合于果蔗种质遗传多样性分析,ISSR标记技术在果蔗遗传多样性分析上比RAPD技术更适用。

【Abstract】 RAPD and ISSR markers were used to assess the germplasm genetic diversity among 40 individuals of chewing cane, including 39 chewing canes comed from different distracts and Badila, 23 RAPD primers and 28 ISSR primers, with polymorphic and informative patterns, were selected from a total of 100 RAPD ones and 100 ISSR ones to determine these individuals’ genetic diversity. The 23 RAPD primers and 28 ISSR primers generated 250 RAPD fragments and 301 ISSR fragments, respectively. For RAPD marker, 175 polymorphic bands were produced, percentage of polymorphic bands was 70%; For ISSR marker, 232 polymorphic bands were produced, percentage of polymorphic bands was 77.1%. Jaccard’s genetic similarity matrices and dendrograms for the 40 individuals were formed based on two molecular markers data. In dendrograms, these chewing canes could be divided into four groups: the first group which was proved to be hybrid species contained five individuals such as Baishan, Waigandan, Dudu, wenling chewing canes and artificial hybrid species 474; the second group was Badiia and Fengcengzipichewing cane; the third group only contained one species------Huangpichewing cane; the fourth group was composed of 32 individuals which comed from different provinces such as Fuji, Jianxi, Guizou, Yunnan, Guangxi and so on. The correlation coefficient was 0.9471 between RAPD and ISSR markers. The results revealed that RAPD and ISSR markers were suitable for assessment of germplasm genetic diversity of chewing cane, and ISSR marker was superior to RAPD marker.
