骨髓间充质干细胞向骨细胞诱导分化及其组织工程化骨组织的实验研究

【摘要】 目的:骨组织工程通过成骨细胞的诱导分化与增殖,并在体内或体外生长活体骨组织,用于骨的修复和再生,种子细胞和基质材料是组织工程研究的主要内容。间充质干细胞(mesenchymal stem cells,MSCs)是一种具有多向分化潜能的细胞,在特定的条件下可向成骨细胞、软骨细胞、肌腱、脂肪细胞、骨髓基质细胞、血管内皮细胞及神经细胞样细胞分化,且易于获得及扩增,是骨组织工程研究中常用的种子细胞。基质材料是细胞附着的基本框架和代谢场所,其形态和功能直接影响着所构成的组织的形态和功能。本实验的目的是研究骨髓间充质干细胞和纳米材料在构建组织工程化骨组织以及修复骨缺损中的作用与特点。 方法:首先分离人骨髓间充质干细胞(bone marrow mesenchymal stem cells,BMSCs),通过常规培养,在体外观察其生长特性、抗原表达等特征;然后通过条件培养基定向诱导,观察其分化潜能;其次分离培养兔骨髓间充质干细胞,与纳米晶羟基磷灰石胶原材料(nHAC)于体外联合培养,通过大体观察、组织学分析及电镜观察了解成骨情况;最后建立兔桡骨缺损模型,植入组织工程骨,通过大体观察、组织学分析及X光摄片观察骨缺损修复情况。 结果:人骨髓间充质干细胞在体外可以大量增殖,常规培养条件下,原

【Abstract】 Objective: The bone tissue engineering repair and regenerate bone through osteoblast generation and induce differentiation as well as growth living bone tissue in vivo vitro. The seeds cell and base material is the main investigative content of tissue engineering. Mesenchymal stem cells is a kind of multi-directional differentiation potency cell and can differentiate into osteoblast、 chondrocyte、 tendon、 adipocyte、 marrow stroma cell、 vascular endothelial cell and nerve cell under specific condition, it was general all purpose seeds cell in bone tissue engineering investigate also because of easy to obtain and generation this cell. The base material is the basic framework and metabolism site of adherent cell, it form and function have direct influence on the tissue form and function construct by itself. The objective of this study is to investigate the contribution and characteristic of bone marrow mesenchymal stem cells as seed cell and the nanometer crystal hydroxyapatite collagen material as scaffold material in bone tissue engineering also in repairing bone defect.
