含连三唑基的1,5-苯并硫氮杂卓,1,5-苯并二氮杂卓及吡唑啉类衍生物的合成

【摘要】 1,5-苯并硫氮杂卓和1,5-苯并二氮杂卓(简称苯并杂卓)是一类具有重要的药理活性和生理活性的七元杂环化合物。多年来,1,5-苯并杂卓类杂环化合物的结构,合成及其生理活性的研究受到国内外的广泛重视。其活泼的亚胺(C = N)双键的环加成反应引起了化学家们的极大兴趣,因为该官能团是对杂卓进行化学修饰和衍生化的主要反应部位之一,由此出发可合成和开发一系列新的1,5-苯并杂卓类衍生物;吡唑啉衍生物是良好的光致发光材料,同时吡唑啉环也是构成不同生物活性的农药的重要结构单元,1,2,3-三唑基团本身也具有许多的生理活性,因此将连三唑基引入到1,5-苯并硫氮杂卓,1,5-苯并二氮杂卓和吡唑啉分子中,可能可以增强其生理活性。本论文的工作分为以下两大部分:第一大部分为文献综述部分,文中对1,5-苯并硫氮杂卓和1,5-苯并二氮杂卓及吡唑啉类荧光化合物的合成、化学改造等在国内外的研究状况作了较为系统的阐述。第二大部分为实验部分,主要的工作为:1.含2-苯基-1,2,3-三唑基的1,5-苯并硫氮杂卓衍生物的合成将(对位取代)苯甲醛与(对位取代)苯乙酮发生羟醛缩合,得到十五种查尔酮,以此为原料与邻氨基苯硫酚反应合成出1,5-苯并硫氮杂卓,然后再用1,5-苯并硫氮杂卓与含2-苯基-1,2,3-三唑取代基的氧化腈偶极子发生1,3-偶极环加成反应,得到含2-苯基-1,2,3-三唑基的1,5-苯并硫氮杂卓衍生物。2.含2-苯基-1,2,3-三唑基的1,5-苯并二氮杂卓衍生物的合成以葡萄糖为基础原料,经过成脎,成环及氧化断链等步骤制得三唑醛(或溴代三唑醛),将其与对位取代苯乙酮(或苯乙酮)发生羟醛缩合,合成出十种查尔酮,用此查尔酮为原料制得1,3-二酮化合物,将其与邻苯二胺反应得到1,5-苯并二氮杂卓。再用苯并二氮杂卓在室温条件下进行1,3-偶极环加成反应,得到1,5-苯并二氮杂卓衍生物。

【Abstract】 1,5-Benzothiazepines and 1,5-Benzodiazepines are a kind of seven- membered ring heter- ocyclic compounds which possess many pharmacology activities and biological activities. For many years, their structure, synthetic mothods and biological activities attracted the extensive attention both at home and abroad. The ring addition reaction of the C=N double bond has caused the chemists’ great interest, because this functional group carries on one of the main reactive position of chemical modification and derivatization. Therefore, we can synthesize and develop a series of the new 1,5-Benzothiazepine and 1,5-Benzodiazepine derivatives; The pyrazolline derivatives is the good materials of photoluminescence, at the same time the pyrazolline ring is the important structure unit of the pesticide whice have different biological activation too; 1,2,3-triazole ring have many pharmacology activation in itself, Therefore, we expect to realize reinforcement of many physiological activities by mean of combining the new group, 2-phyenyl-1,2,3-trizole which have strong biological activities with 1,5-Benzothiazepine and 1,5-Benzodiazepine and pyrazolline.The thesis is includes two major parts:The first part is about literature review. The important synthetic methods, the structure and chemical transformation of 1,5-Benzothiazepine and 1,5-Benzodiazepine and pyrazolline derivatives that have studied in home and other countries were explicated systematically.The second part is the experiment part, the main work as follows:1. Synthesis of 1,5-Benzothiazepine Derivatives containing 2-Phenyl-1,2,3-triazole. phenyl aldehyde (or para-substituted phenyl aldehydes) reacted with acetophenone (or para-substituted acetophenones) to form fifteen chalcones (1a~1o) by aldol condensation.
