中药复方免疫增强剂中活性多糖的分离纯化研究
Study on Separation, Purification of Bio-active Polysaccharides in a Chinese Herbal Medicine-Compound Immunoenhancer

【摘要】 因为在抗肿瘤、抗病毒、降血糖、抗炎、抗衰老和抗凝血等方面发挥着重要的作用,多糖成为科学研究的热点之一。本研究提取了复方免疫增强剂中的多糖,对多糖进行了分离纯化,确定了所得3个多糖均一组分的相对分子质量以及单糖组成。1.采用超声辅助法提取了复方免疫增强剂中的多糖,并采用苯酚-硫酸法进行了多糖含量测定的研究。实验结果显示,多糖浓度在24.2~96.8μg/mL内与吸光度线性关系良好,相关系数R=0.9994。同时本文采用此法对复方免疫增强剂多糖提取物进行了含量测定,测得多糖含量为42.82%;2.采用乙醇分级沉淀、DEAE-纤维素层析及Sephadex G-200凝胶色谱等方法对多糖进行了分离纯化研究,最终得到复方免疫增强剂中三个主要多糖组分PS1-A-Ⅰ、PS2-A-Ⅱ及PS3-A-Ⅲ;3.运用Toyopearl HW-55F凝胶色谱法、纸层析法和紫外分光光度法进行了纯度鉴定,证明PS1-A-Ⅰ、PS2-A-Ⅱ及PS3-A-Ⅲ均为多糖均一组分,不含有核酸和蛋白质;4.用Toyopearl HW-55F凝胶层析测得PS1-A-Ⅰ、PS2-A-Ⅱ及PS3-A-Ⅲ的相对分子质量Mr分别为389,941、124,451、7,586;通过完全酸水解、纸层析法初步确定了三者的单糖组成:PS1-A-Ⅰ是由葡萄糖构成的均多糖,PS2-A-Ⅱ由阿拉伯糖、甘露糖和半乳糖构成,PS3-A-Ⅲ是由阿拉伯糖和半乳糖构成的杂多糖。本研究从提取物中分离出3种多糖均一组分,为探讨该中药复方免疫增强剂的免疫机理提供支持,同时本研究将促进新疆本土药用植物资源的利用及中药免疫增强剂研究,并为进一步研究CIE中活性多糖的免疫机理、开发新型高效的畜禽免疫增强剂打下了基础。
【Abstract】 Because it’s avidity on antitumour, antivirus, anticaducity, antiinflammation, anticruor and decreasing blood sugar et al, polysaccharide has become a hot field of scientific studies. In this paper, polysaccharides in Compound Immunoenhancer were extracted and separation was carried out towards active polysaccharides. Three polysaccharides were obtained with their relative molecular weights (Mr) and their monosaccharide compositions confirmed.1.The polysaccharides in Compound Immunoenhancer were extracted adopting ultrasonic-assisted extraction method and content determination was done applying the method of phenol-sulphate acid. Results of content determination showed that the calibration curve of polysaccharide was linear with absorbance in the range from 24.2μg·mL-1 to 96.8μg·mL-1, with R= 0.9994. This method was next used to measure the content of polysaccharide in Compound Immunoenhancer, which proved that the content of the extract was 42.82%.2.Three polysaccharides, PS1-A-Ⅰ, PS2-A-Ⅱa nd PS3-A-Ⅲ,were obtained adopting ethanol-precipitating, DEAE-Cellulose ion-exchange chromatography and Sephadex G-200 gel chromatography.3.Three polysaccharides were proved homogeneous by TOYOPEARL HW-55F gel chromatography, paper chromatography and ultraviolet spectrophotometry. And no nucleic acid and protein were found.4 . The relative molecular weights of three homogeneous polysaccharides were determined 389941, 124451, and 7586 by TOYOPEARL HW-55F gel chromatography. The monosaccharide composing three polysaccharides were also investigated: PS1-A-Ⅰwas composed of one kind of sugar——glucose, PS2-A-Ⅱof galactose, mannose and arabinose, and PS3-A-Ⅲof two kinds——galactose and arabinose.Three polysaccharides got in this study provided support for investigation on immune mechanism of this Chinese herbal medicine. At the same time, this paper would promote the utilization of officinal resource in Xinjiang and studies on traditional Chinese medicine which enhances immune system, and it’s sure to be helpful to researches on the mechanism of Compound immunoenhancer.
