有机染料分子和DNA相互作用的电化学研究及分析应用

【摘要】 DNA作为遗传信息的载体,是生命活动中一类重要的生物大分子,研究各种小分子如抗癌药物、金属配合物和染料分子与DNA的相互作用在致癌机理、药物筛选和疾病诊断方面有着重要意义。本文筛选了具有电活性的染料分子吡啰红B、酚藏花红、灿烂甲酚蓝、结晶紫和中性红作为电化学探针,采用电化学法在汞电极上系统研究了它们的电化学行为以及和DNA的相互作用。 论文由6章组成,第1章综述了小分子和DNA的相互作用;第2至6章分别筛选了吡啰红B、酚藏花红、灿烂甲酚蓝、结晶紫和中性红等小分子染料,以汞电极如静汞电极、滴汞电极作为工作电极,采用循环伏安法、线扫极谱法等电化学手段,结合紫外-可见分光光度法,研究了它们的电化学行为以及和DNA的相互作用。通过对加入DNA前后峰电流、峰电位以及一些电化学参数如电子转移系数α、标准速率常数K_s和扩散系数D_o的研究发现,这些小分子均可以与DNA发生结合反应导致染料分子的峰电流降低。利用峰电流的变化对结合反应的条件和电化学检测条件进行了优化,考察了常见的金属离子、氨基酸等物质的存在对结合反应的影响。在最佳条件下,峰电流的降低值同DNA浓度在一定范围内成良好的线性关系,进而可用于DNA的分析,建立了测定DNA的工作曲线,应用于混合样品的测定,获得了令人满意的结果。利用电化学法求解了反应的结合比和结合常数,对结合反应的模式进行了推导。

【Abstract】 DNA is one of the most important biomacromolecules in life processes as the carrier of genetic information. The studies of some low molecular weight compounds such as anticancer drugs, metal complexes and organic dyes with DNA are crucial fields in mutation detection and clinical diagnostics. In this paper, the electrochemical behaviors of some electroactive dyes such as pyronine B, phenosafranine, brilliant cresyl blue, crystal violet, neutral red etc. and their interaction with DNA on the mercury electrode were carefully studied.This paper is made up of 6 chapters; chapter 1 summarizes the interaction of low molecular weight compounds with DNA; chapter 2 to 6 focus on the electrochemical studies of some organic dyes such as pyronine B, phenosafranine, brilliant cresyl blue, crystal violet, neutral red etc. and their interaction with DNA, respectively. Different electrochemical methods such as linear sweep voltammetry, cyclic voltammetry and UV-Vis spectrophotometric method are applied to the investigation. Based on the changes of their peak currents, potentials and some electrochemical parameters such as the charge transfer coefficient a, the electrode reaction standard rate constant k_s and the diffusion coefficient Do before and after the addition of DNA, it is found that these dyes can bind to DNA and lead to the decrease of reduction peak current. The stoichiometry of these supramolecular complexes such as the binding number and the binding constant are calculated by electrochemical data. The conditions of the binding reaction and the electrochemical determination are optimized. The influences of coexisting substances such as metal ions and amino acids are tested.
