鼻咽癌中STGC3基因的表达及调控其表达CNE2细胞系的建立

【摘要】 第一部分 STGC3蛋白在鼻咽癌组织中的表达及其意义 目的:通过观察STGC3蛋白质在鼻咽癌和正常鼻咽活检组织中的表达及分布情况,探讨STGC3在鼻咽癌发生发展中的作用。 方法:首先制备兔抗人STGC3多克隆抗体;运用免疫组化SP方法,检测40例鼻咽癌组织及21例正常鼻咽组织中STGC3蛋白的表达与定位;运用Western blot法,检测20例鼻咽癌和20例正常鼻咽活检标本中STGC3蛋白质的表达水平。 结果: (1)免疫组化结果:正常鼻咽粘膜组织STGC3蛋白定位于上皮细胞胞核及胞浆,阳性表达率为85.71%(18/21),鼻咽癌中STGC3主要在癌细胞的胞浆表达,胞核表达较少,阳性表达率为30.00%(12/40),明显低于正常对照组织,差异具有显著性,P<0.05:鼻咽癌STGC3阳性表达率在临床Ⅰ-Ⅱ期(43.75%)明显高于Ⅲ-Ⅳ期(20.83%),且无淋巴结转移的鼻咽癌病例其阳性率(38.10%)明显高于有淋巴结转移的鼻咽癌病例阳性表达率(21.05%),P<0.05;(2)Westernblot结果:鼻咽癌STGC3相对表达量(STGC3/actin)明显低于对照正常组织(0.324±0.185;0.978±0.213),P<0.05。 结论:STGC3蛋白在正常鼻咽粘膜和鼻咽癌中定位于细胞的胞核及胞浆。正常鼻咽粘膜和鼻咽癌STGC3的表达水平及定位有差别,鼻咽癌组织中STGC3蛋白的表达水平明显低于正常对照组织,且鼻咽癌中核表达减少。STGC3阳性表

【Abstract】 Objective: To investigate the expression and distribution of STGC3 in nasopharyngeal carcinoma (NPC) and normal nasopharyngeal biopsy samples to provide some acceptable indications about the function of the candidate tumor suppressor STGC3.Methods: The rabbit anti-STGC3 antibody was prepared by immunizing rabbit with a synthetic peptide derived from the COOH-terminal end of STGCC protein. The expression and distribution of STGC3 protein were displayed by the immunohistochemical staining in 40 NPC and 21 normal nasopharyngeal samples; Western blot was used to analyze the expression level of STGC3 protein in 20 NPC and 20 normal nasopharyngeal specimens.Results: The expression and distribution of STGC3 protein were examined in NPC and normal nasopharyngeal tissues by using immunohistochemistry. The expression of STGC3 was located in cytoplasm and nuclear of epithial cell in normal nasopharyngeal biopsy, but chiefly in cytoplasm of tumor cell in NPC. The positive expression rate of STGC3 was 85.71% (18/21) in in normal nasopharyngeal samples, and 30% (12/40) in NPC, and difference was statistically significant between them (P<0.05). In NPC, the positive expression rate in I -II phase was higher than that in III-IV phase (43.75% and 20.83%, respectively), P<0.05.
