PIG11蛋白在人体组织中的分布特征及其高表达与As2O3诱导细胞凋亡关系的研究

【摘要】 1.PIG11蛋白在人体正常组织及其相应肿瘤组织中表达的研究 目的 检测PIG11(p53-induced gene 11)蛋白在人体正常组织与相应肿瘤组织的表达情况,探讨PIG11蛋白在人体正常组织的分布特征,及其在肿瘤发生发展中所起的作用。 方法 利用免疫组化SP法检测PIG11蛋白在319例人体正常组织与相应肿瘤组织中的表达,及其在人胚肝永生化细胞株L-02和三种人肝癌细胞株Bel-7402、SMMC-7721、HepG2中的表达。 结果 PIG11蛋白在肝、胃、肠、乳腺等腺上皮及鳞状上皮组织中明显表达阳性,在正常胃粘膜腺颈部较体部和底部表达降低;而在纤维、脂肪、平滑肌、淋巴组织及脑组织中无明显表达。PIG11蛋白在肝癌、低分化胃和肠腺癌、乳腺癌组织中较相应正常组织均有明显表达下调(p<0.01);在鳞癌组织中,与正常鳞状上皮比较,也有表达下调(p<0.05);在HepG2细胞中的表达较L-02细胞也明显下调(p<0.01)。 结论 PIG11蛋白在人体组织分布有一定的特异性,且与细胞的分化程度有关;其主要分布在肝、胃肠、乳腺等腺上皮及鳞状上皮组织中,而纤维、脂肪、平滑肌、淋巴组织及脑组织中均无明显表达。PIG11蛋白在上皮性肿瘤组织中表达明显下调,其低表达也许与上皮源性的恶性肿瘤发生发展有关。 2.PIG11蛋白高表达与As2O3诱导细胞凋亡关系的研究 目的 选用高表达PIG11细胞株L-02和低表达PIG11细胞株HepG2,研究PIG11表达与三氧化二砷(arsenic trioxide,As2O3)诱导细胞凋亡的关系,以及在此过程中细胞内活性氧(reactive oxygen species,ROS)、p53蛋白表达变化,进一步探讨PIG11基

【Abstract】 Objective: The aim of this study was to investigate the expression of PIG11 (p53-induced gene 11) protein in human normal tissue and corresponding tumor tissue, discussing the distributing characteristic of PIG11 protein in human normal tissue and its function in the tumorigenesis and development.Methods: The expression of PIG11 protein was determined by immunohistochemical peroxidase-conjugated streptavidin (SP) method in 319 cases of human normal tissues and corresponding tumor tissues. And its expression was determined by the same way in human embryo hepatocytes L-02 and three human hepatocellular carcinoma cell lines Bel-7402, SMMC-7721 and HepG2.Results: It was positive expression of PIG11 protein in human liver, gastric mucosa, intestine mucosa, galactophore glandular epithelium and squamous epithelium tissue. Expression of PIG11 protein in cervical part of normal gastric mucosa gland was lower than that of its caudomedial part and bottom. Its expression was negative in human fibre, fat tissue, smooth muscle, lymph tissue and brain tissue. Expression of PIG11 protein was descent in liver cancer, poorly differentiated stomach adenocarcinoma, poorly differentiated intestine adenocarcinoma, breast cancer, comparing with those of corresponding normal tissues (p<0.01), and its expression was low in squamous carcinoma too, comparing with normal squamous epithelium (p<0.05). At the same time, the PIG11 protein of HepG2 cell line was down-regulated, contrasting with L-02 cell line (p<0.01).Conclusion: There was tissues specificity to the distribution of PIG11 protein in human normal tissues, and its expression was involved in cellular differentiation. It was distributed mostly in human epidermic tissues, including liver, gastric mucosa, intestine mucosa, galactophore glandular epithelium, squamous epithelium tissue, and so on, while, it was negative in human fibre, fat tissue, smooth muscle, lymph tissue and brain tissue.The
