补髓生血颗粒对再生障碍性贫血模型大鼠骨髓基质细胞的影响及长期毒性

【摘要】 目的:研究补髓生血颗粒对正常大鼠的长期毒性及促进再障大鼠骨髓造血的作用和机制。 方法:(1)正常Wistar大鼠给予不同剂量补髓生血颗粒连续灌胃180天后,测定其血液生化学指标,解剖后对重要脏器分别称重,计算脏器系数,并进行病理组织学检查。(2)Wistar大鼠用60Co-γ射线8.0Gy照射造成再障模型,分别给予低、中、高剂量补髓生血颗粒,连续给药3周后,采用集落培养的方法培养骨髓成纤维细胞集落(Colony-forming unit-fibroblast,CFU-F)观察补髓生血颗粒对再障大鼠造血祖细胞集落形成的影响。 结果:(1)连续用药180天后,补髓生血颗粒低、中、高剂量组大鼠多数血液生化学指标与正常对照组比较无明显差异,仅个别指标有轻度变化,但停药15日后可迅速恢复;各组大鼠之间脏器系数无明显差异;低、中剂量补髓生血颗粒对心、肝、脾、肺、肾、胃、脑等重要脏器无明显影响,高剂量补髓生血颗粒可导致心脏、肾脏、肝脏有瘀血、出血、水肿轻微病理变化,但此种变化是可逆的。(2)体外集落培养,低、中、高剂量补髓生血颗粒使CFU-F集落产量提高,与模型对照组比较有显著差异(P<0.05),与再障生血片组比较亦有显著差异(P<0.05);病理切片可见造血组织增生,分布较均匀,出现造血组织基本结构修复现象。 结论:补髓生血颗粒对正常大鼠血液生化指标及脏器系数无显著影响,对大鼠各重要脏器无明显毒副作用。补髓生血颗粒能够促进再障大鼠骨髓CFU-F集落的形成,刺激造血祖细胞的增殖,修复造血组织基本结构,从而改变再障大鼠骨髓抑制状态,改善大鼠生存质量,补髓生血颗粒对再障大鼠骨髓造血的促进作用优于再障生血片。

【Abstract】 Objective: To investigate the long term toxicity of Marrow-Supplementing and Blood-Engendering Granule for normal Wistar rats and the effects of Marrow-Supplementing and Blood-Engendering Granule on bone marrow and peripheral blood of Wistar rats with aplastic anemia.Method: (1) The biochemistry of normal Wistar rats were observed and the key organs were cut for calculating the viscera coefficient and pathological examination after being treated with Marrow-Supplementing and Blood-Engendering Granule at higher, middle and low dosages for 180 days. (2)After shined on the 60Co-Y ray with 8.0Gy, Wistar rats caught aplastic anemia and were treated with Marrow-Supplementing and Blood-Engendering Granule at higher, middle and low dosages for three weeks, The colony formation of CFU-F of Wistar rats in aplastic anemia were observed by semi-solid culture of bone marrow.Results: (1)After given medicine for 180 days, the groups treated with Marrow-Supplementing and Blood-Engendering Granule had no significant difference in biochemistry data, and only very few light changes could recover without the medicine in 15 days;the viscera coefficient of all groups had no significant difference;the low and middle dosage had no significant influence to major viscera and the higher dosage led to light changes of blood clot, bleeding and edema within heart, kidney and liver, but the changes were deteriorated. (2)The low, middle and higher dosage medicine could make the productivity of CFU-F cultured outside the body. It was a significant difference to compare with the model group and the group with the zaizhangshengxue tablet (P<0.05);it could be seen hematopoietic tissue was multiplied,well-distributeand recovery when the tissue slices of pathology were observed.Conclusion: Marrow-Supplementing and Blood-Engendering Granule had no side-effect or significant influence to biochemistry and viscera coefficient. It could promote the CFU-F forming, stimulate HSC multiplying and recover the hematopoietic tissue, so it-could change the state of bone marrow inhibition of Wistar rats caught aplastic anemia and improve their living quality. Marrow-Supplementing and Blood-Engendering Granule showed more effective than zizhangshengxue tablet.
