嗅成鞘细胞移植治疗皮层脑梗塞的实验研究

【摘要】 背景和目的 成熟哺乳类动物中枢神经系统受到诸如缺血等原因的损伤后由于缺血中心区大量神经元死亡,造成严重的功能障碍。已有研究表明脑梗塞后梗塞灶周围皮层神经元轴突向梗塞灶生长,并形成新的突触联系,这与行为功能的恢复有关。中枢神经损伤之后,再生能力较低的重要原因是由于中枢神经损伤后的胶质细胞增生和各种抑制因子的存在。嗅成鞘细胞(Olfactory ensheating cells,OECs)是一种十分独特的细胞,由于具有一定的神经再生的“容许性”(Permissiveness),能够分泌多种神经营养因子,具有促进中枢神经再生的能力。已有实验证实OECs移植能促进脊髓损伤神经纤维的再生并促进其功能的恢复。但是,它在脑梗塞中的治疗作用未有报导。本研究建立了培养、纯化OECs的方法并观察OECs移植后在体内存活情况,进一步探讨OECs细胞对大鼠脑梗塞后功能恢复的影响。材料与方法 一、细胞培养 从成年SD(Sprague-Dawley,SD)大鼠嗅球(Olfactory Bulb,OB)取材进行OECs原代培养。利用差异贴壁方法进行纯化培养,清除绝大部分神经元及其它混杂细胞,使细胞成分相对单一化。低亲和力神经生长因子受体(low affinity growth factor receptor,L-NGFR)免疫染色鉴定OECs,移植前,双苯亚甲胺
【Abstract】 Background and ObjectiveTraumatic injuries such as ischemia to the adult mammalian central nervous system (CNS) cause severe disabilities, because of the loss of large number of neurons in the core of infarct. Some researches suggest the involvement of axonal sprouting followed by synaptogenesis in the neocortex after focal ischemia as a mechanism for function recovery. Some important factors for lack of regeneration in the central nervous system after traumatic injuries are the extensively existent of nerve growth inhibitors and gliosis.Olfactory ensheathing cells (OECs) are very unique cells and hypothesized to facilitate regeneration of central nervous system and excreting neurotrophic factors.It has been proved that olfactory ensheathing cells (OECs) transplantation on the spinal cord injuries can promote the regeneration of spinal cord axons and functional recovery, but there was no research on the OECs transplantation in cerebral infarction. Our experiment establishes a simple and efficiency method for OECs culture and purification , OECs survival and repair in vivo for cerebral infarction after the OECs transplantation were also observed.
