脑肿消对缺血性中风急性期脑水肿的实验研究

【摘要】 目的:通过中药复方脑肿消干预大鼠局灶性脑缺血后脑水肿的实验研究,探讨分析脑肿消围攻缺血性中风急性期脑水肿的作用机制,为其临床应用提供实验依据。方法和内容:采用线栓法建立大鼠局灶性脑缺血模型,观察大鼠神经功能行为学改变并进行评分,检测脑组织含水量、脑组织钠离子(Na+)、钾离子(K+)、钙离子(Ca2+)、一氧化氮(NO)含量,测定钠钾三磷酸腺苷酶(Na+-K+-ATPase)及钙三磷酸腺苷酶(Ca2+-ATPase)活性。结果:脑肿消大、小剂量组及维脑路通组均可不同程度地降低脑组织Na+、Ca2+、NO的含量,升高K+含量、Na+-K+-ATPase及Ca2+-ATPase活性,与模型组比较,差异均有统计学意义(P<0.05),且以脑肿消大剂量组作用最显著。结论:脑肿消对大鼠局灶性脑缺血后脑水肿有防治作用,其机制可能与改善能量代谢障碍、调节细胞内外离子平衡、降低脑内NO水平有关,其它药理机制有待于进一步探讨。
【Abstract】 Objective: To ascertain the pharmacological effect of traditional Chinese medical compound naozhongxiao that can intervene the cerebral edema in rats with focal cerebral ischemia. And to probe and analyse the action mechanism of naozhongxiao in many ways in cerebral edema at the acute ischemic apoplexy stage. The results of the study provided an experimental basis for developing the clinical application of naozhongxiao. Methods and Contents: It had been erected that the model of focal cerebral ischemia with suture-occluded method in rats in order to observe the effects of naozhongxiao on the content of cerebral water, Na+, K+, Ca2+, nitric oxide (NO) and the activity of Na+-K+-ATPase, Ca2+-ATPase. Results: Both naozhongxiao and weinaolutong could decrease to some degree the content of cerebral water, Na+, Ca2+ ,NO (P<0.05) and increase the conten of K+, the activity of Na+-K+-ATPase, Ca2+-ATPase (P<0.05). Compared with model group, the high dose group of naozhongxiao showed most significant effect. Conclusion: Naozhongxiao showed markedly preventive effect on cerebral edema in focal cerebral ischemia rats. The mechanism could be concerned with relieving the energy dysmetabolism, acommodating the ionic balance of exterior and interior of cell and decreasing the NO level in brain. The other pharmacological mechanisms of naozhongxiao have yet to be Investigated.
