兔源致病性大肠杆菌OMP_s差异性分析及主要OMP免疫保护性的初步研究

【摘要】 随着我国养兔业的发展,家兔的疫病流行也呈现出新规律,其中家兔腹泻尤其是大肠杆菌性腹泻,已成为仔幼兔生长受阻和死亡率高的重要原因之一。本研究在对兔源致病性大肠杆菌进行细菌分离鉴定的基础上,再对该地区的兔源致病性大肠杆菌分离株的OMP及其分型进行系统的研究,以便了解该地区的兔源致病性大肠杆菌分离株的遗传相关性,给防治本病提供依据;进一步通过动物实验和细胞水平的实验,研究代表菌株外膜蛋白在体液和细胞免疫水平上诱导实验动物免疫保护的效果。为下一步研制临床免疫制剂和疫苗提供了一定基础,必将会大大推动兔大肠杆菌病的诊断和防治工作的进步,有利于兔养殖业的发展。本研究共分三部分:1.兔源致病性大肠杆菌的分离鉴定本研究2003,2004,2005年连续三年从潍坊某地区的7个兔场采取典型腹泻病、死兔的肝脏和各段肠管的内容物以及健康兔粪便或直肠拭子做为备检材料共152份。通过常规的方法分离鉴定方法,共鉴定出典型大肠杆菌共166(2003年为47株,2004年为68株,2005年为51株)株,通过致病性试验初步鉴定出兔源致病性大肠杆菌共为96株(2003年为28株,2004年为46株,2005年为32株),并选择代表菌株做了药物敏感试验。研究内容包括流行病学、临床表现、病理形态学、病原形态及理化特征、药敏试验等,从中发现了一些规律和特点,填充了家兔大肠杆菌病的研究内容,对有效地诊断和防治家兔大肠杆菌病具有一定的指导意义。2.致病性大肠杆菌外膜蛋白型的确立及差异性分析提取从该地区分离并鉴定为致病性大肠埃希氏菌的菌株的OMP,经SDS-PAGE电泳检测外膜蛋白,确立其外膜蛋白型(OMPs),衡量不同菌株
【Abstract】 Along with the development of rabbit industry, the rabbit diarrhoea disease was also serious increasingly, especially the diarrhoea caused by E.coli, The young rabbit precede-weaned or post- weaned had a high death rate, leading to a heavy economy lose, thus was seriously restricting the growth of young rabbit development of rabbit industry. This research was based on the isolation and identification of pathogenic E.coli from rabbit, and their OMPs were determined and their genic association was weighed, which provide thereunder for the prevention and cure of E.coli. And then and then Protective Immunity of Pathogenic Escherichia Coli Isolates Of Rabbits Origin were studied on the level of animal and cell,which provided theory basis for studying falling ill mechanism and the control measure of this disease and provide a quick and accurately diagnosis for rabbit diarrhoea. This study consists of three parts.PartⅠIsolation and identification of pathogenic rabbit Escherichia coliIn this section, 152 shares of samples isolated from rabbits in this region in 2002, 2003, 2005 were isolated and identificated, 166 strains (47 strains in 2003,68 strains in 2004,51strains in 2005)of Escherichia coli of rabbit were obtained.
