嗜热毛壳菌纤维酶对小尾寒羊瘤胃发酵及粗饲料降解的影响

【摘要】 本论文采用体外法与体内法相结合的研究方法,研究新型酶制剂—嗜热毛壳菌纤维酶对粗饲料降解效果及对小尾寒羊瘤胃发酵的影响,探讨其在绵羊生产应用中的有效性。主要的研究结果如下:1在20℃、40℃和60℃温度下,研究嗜热毛壳菌纤维酶对玉米秸纤维物质降解效果,结果表明:1)不同温度显著影响酶对玉米秸纤维物质的降解效果(P <0.05),60℃优于40℃和20℃;2)酶的剂量也影响其降解效果(P <0.05),0.9%水平组优于0.3%和0.6%水平组。2从4只安装有永久性瘤胃瘘管的小尾寒羊(45±0.5 kg)中采集瘤胃液处理嗜热毛壳菌纤维素酶,用体外法研究其稳定性。试验结果表明,该酶在瘤胃液中有较好稳定性。3选用4只安装有永久性瘤胃瘘管的小尾寒羊(45±0.5 kg),采用4×4拉丁方试验设计,通过体内法和尼龙袋法研究不同水平嗜热毛壳菌纤维酶对小尾寒羊瘤胃代谢和粗饲料降解率的影响。试验结果表明:1)添加酶使瘤胃VFA浓度显著提高(P <0.05),pH显著下降(P <0.05),且以0.6%水平组较好,但对瘤胃NH3-N浓度影响不显著;2)日粮中添加酶显著提高玉米秸和麦秸纤维素的降解率(P <0.05);3)不同种类的粗饲料在瘤胃中的降解率不同,在相同酶水平下玉米秸在瘤胃中的降解率大于麦秸。4选用80只健康的小尾寒羊来研究不同水平纤维酶对小尾寒羊生产性能的影响。试验结果表明:绵羊日粮中添加酶能显著提高总增重和日增重及NDF、ADF、CEL及HC的消化率(P <0.05),在本试验中以添加0.5%水平的酶效果较好。综合以上的试验结果表明,嗜热毛壳菌纤维酶能够提高粗饲料的降解率和饲料养分利用率,改善反刍动物生产性能。

【Abstract】 This paper mainly studied the feeding responses to C. thermophile fibre enzyme in sheep and the degradation of coarse fodder in vivo and in vitro. By four tests, the article proved that the enzyme has action in sheep. The results showed:1. The effect of the cellulase complex on degradation of fibre in the corn stovers at 20℃, 40℃and 60℃were studied. The results showed, (1) The different temperature had significantly effected on the degradation of fibre(P <0.05), which was better at 60℃than at 20℃or 40℃; (2) The effect on the degradation of fibre was different with the dosage of the enzymes(P <0.05), which was better for the level of 0.9% than the level of 0.3% and 0.6%.2. Four Small Tail Han-sheep with average weight of 45kg fitted with permanent ruminal fistulae were used to study the stability of C. thermophile cellulase. C. thermophile cellulase was treated in vitro by ruminal liquid of sheep. The results indicated that the cellulase was stabler.3. Four Small Tail Han-sheep with average weight of 45kg fitted with permanent ruminal fistulae were used to study the effects of C. thermophile cellulase on rumen fermentation and the degradation of coarse fodder by vivo and nylon bag technique. The experiment was performed in 4×4 Latin square design. The results indicated that, (1) the VFA concentration was significantly increased(P <0.05), pH values were reduced(P <0.05), which was better for the level of 0.6%; but the ruminal ammonia-N concentration had no effect; (2) the enzyme significantly increased the degradation of fibre in the corn stover and wheat stalk(P <0.05); (3) the different types of coarse fodder have different degradations in the rumen of sheep, which was better for the corn stover than the wheat stalk in the same level of the enzyme.4.
