ICT荧光探针与生物大分子相互作用的研究

【摘要】 第一章:本章简要概述了分子内电荷转移荧光的原理及研究现状、蛋白质光谱探针的研究进展和DNA光谱探针的研究进展。 第二章:利用吸收光谱和荧光光谱对化合物:4’-二甲氨基-2,5-二羟基查尔酮(DMADHC)在不同极性介质中的光谱、光物理行为进行了研究,并利用Lippert-Mataga方程计算了该化合物基态和激发态偶极矩间的差值。实验结果表明,溶剂极性使DMADHC的吸收光谱产生相对较小的红移现象,而使其荧光光谱发生较大的红移,且基态和激发态间的偶极矩差值△μ较大,证明了该化合物具有典型的分子内电荷转移(ICT)的特性。 第三章:本章采用紫外可见和荧光分光光度法研究了一种新型的分子内电荷转移荧光探针1-酮-2-(对二甲氨基苯亚甲基)-四氢萘在不同极性有机溶剂中的发光行为和光物理性质。随着溶剂极性的增加,吸收光谱出现轻微红移,而荧光光谱出现显著的红移并伴随着荧光峰的半高宽变宽,量子产率出现先增加后减小的趋势。用Lippert-Mataga方程计算了该化合物基态和激发态偶极矩间的差值。结果证明该化合物在激发态存在着强烈的分子内电荷转移行为。 第四章:本文研究了新型的分子内电荷转移荧光探针4’-二甲氨基-2,5-二羟基察尔酮与人血清蛋白的相互作用。随着人血清蛋白含量的增加,探针的荧光强度显著增大,荧光峰位置逐渐蓝移。同时,与蛋白的结合也使得探针的吸收光谱发生红移,并出现一个明显的等吸收点。通过Scatchard方程得出探针和HSA的结合常数和结合位点数,并计算了结合反应过程的热力学参数,确定了探针和HSA的主要作用方式,并且对可能的结合位点进行了推测。此外,实验探讨了影响探针与人血清蛋白作用的条件,并建立了一种新型、简便的人血清蛋白的检测方法。对HSA检出的线性范围为1—95μg/mL,检出限为0.5μg/mL。应用本方法测定人

【Abstract】 Chapter 1: The principle and the development of intramolecular charge transfer fluorescence were described. Recent developments of the HSA and DNA interactions with Spectroscopic probe were been reviewed.Chapter 2: The spectrum and photophysical properties of a new intramolecular charge transfer (ICT) probe namely 4’-dimethylamino-2,5-dihydroxychalcone (DMADHC) were studied in different solvents by using steady-state absorption and emission spectroscopy. Whereas the absorption spectrum undergoes minor change with increasing polarity of the solvents, the fluorescence spectrum experiences a distinct bathochromic shift in the band position and the fluorescence quantum yield increases reaching a maximum before decrease with increasing the solvent polarity. The magnitude of change in the dipole moment was calculated based on the Lippert-Mataga equation. These results give the evidence about the intramolecular charge transfer (ICT) character in the emitting singlet state of this compound.Chapter 3: The photophysical properties of a new compound 1-Keto-2-(p-Dimethylaminobenzal)-Tetrohydronaphthalene in various solvents at room temperature were characterized by the absorption and steady-state fluorescence technique. The bathochromic shift on the emisson spectra, the broad halfwidth of the fluorescence band and the increase in the excited state dipole moment occurred. These gave the evidence about the intramolecular charge transfer (ICT) character in the emitting singlet state of the compound.Chapter 4: 4’-dimethylamino-2,5-dihydroxychalcone (DMADHC) exhibited dramatic enhancement of fluorescence intensity with accompanying blue-shift of the emission maximum when the concentration of human serum albumin (HSA) was increased.
