自然发酵酸马奶中乳杆菌的DNA多态性研究

【摘要】 本研究以分离自蒙古国和内蒙古锡林郭勒盟等地区自然发酵酸马奶中的51株乳杆菌为材料,采用ERIC-PCR和AFLP两种分子标记方法进行DNA多态性研究。根据AFLP和ERIC-PCR扩增条带,通过UVP-labworks软件,将AFLP和ERIC-PCR扩增结果转换为数据,采用NTSYS软件统计并分析后,生成相应的UPGMA分类树。以相似性系数0.80为分类依据,共分为11个ERIC类群和8个AFLP类群。其中,AFLP标记不能很好的将L. coryniformis subsp. coryniformis、L. kefiranofaciers与L. acidophilus区分;而ERIC-PCR指纹图谱分类结果与传统分类鉴定结果吻合。在此基础上,进一步对15株L.casei、17株L. acidophilus以及2株标准菌株进行种内分型。同样以相似性系数0.80为分类依据,L.casei分为6个ERIC基因型和7个AFLP基因型;L. acidophilus分为12个ERIC基因型和11个AFLP基因型。2种标记技术分类的结果表明,ERIC-PCR和AFLP标记在种内分型大部分不吻合,但菌株来源不同均表现为不同的ERIC基因型和AFLP基因型。
【Abstract】 51 strains of Lactobacillus were pure culture isolated from traditional home-made koumiss collected in the Mongolia and XiLinGuoLe region, Inner Mongolia, china. The DNA polymorphism of the use of afternative molecular markering methods were investigated:enterobacterial repetitive intergenic consensus PCR (ERIC-PCR) and amplified fragment length polymorphism (AFLP). All achieved results from 2 applied markering methods were analyzed with the UVP-labworks software. The data were used to construct UPGMA dendrograms with the NTSYS software. All strains can be classified into 11 ERIC and 8 AFLP groups (similarity coefficient = 0.8). Strains of L. coryniformis subsp. coryniformis, L. kefiranofaciers and L. Acidophilus could not be distinguished efficiently by AFLP. The ERIC-PCR results were consistent with that of the traditional method.In the further study, genotype identification in genus of 15 L.casei strains, 17 L. acidophilus and 2 standard strains were carried out. The result showed that L.casei can be classified into 6 ERIC and 7 AFLP genotypes, L. acidophilus can be divided into 12 ERIC and 11 AFLP genotypes (similarity coefficient = 0.8). The results of two markering methods indicated that most of the ERIC-PCR and AFLP classifications results were not inosculation, but the strains isolated from diffierent area exhibited different ERIC and AFLP genotype.
