不同季节牛初乳对新生犊牛被动免疫影响的研究

【摘要】 本论文采用高效液相色谱法测定了牛初乳中IgG含量。并在此基础上比较了牛初乳中常规成分和IgG含量的季节性变化规律和变化幅度,同时对采食不同季节牛初乳的新生犊牛的血清成分(IgG、IgA、IgM、IGF-I、INS、T3、T4)进行了检测,对犊牛发病率和死亡率进行了统计,进而探讨了不同季节牛初乳对新生犊牛获得性免疫是否有显著影响。试验结果如下:与琼脂双扩散法相比,高效液相色谱法具有分析速度快、灵敏度和精确度高、平均回收率高、分离效果好、所需样品用量少等优点。夏季牛初乳中168h内的蛋白质、IgG、非脂乳固体、密度、pH值;32h内的总干物质含量;64h~168h内的乳糖含量均高于冬季牛初乳相对应时间内的相对应成分;而168h内的脂肪含量、48h内的乳糖含量、48h~168h内的总干物质含量均低于冬季牛初乳相对应时间内的相对应成分。其中除2h内IgG、32h内蛋白质、32h后脂肪含量、64h内密度差异显著外(P < 0.05),其余指标差异均不显著(P > 0.05)。冬季犊牛血清中各成份含量在犊牛采食后144h内均高于夏季犊牛血清中相对应时间内的相对应成分。其中IgG、T3、T4差异变化显著(P < 0.05),IgA、IgM、IGF-I和INS差异变化不显著(P > 0.05)。犊牛血清中高浓度的IgG、IgA、IgM、INS、IGF-I、T3和T4对新生犊牛早期肠道成熟和生长发育具有重要作用。不同季节牛初乳成分显著影响新生犊牛的被动免疫(P < 0.05)。

【Abstract】 The content of immunoglobulin G in bovine colostrums was measured with high performance liquid chromatograph in this paper. Then the changeable regulation of the routine components and IgG in different seasonal colostrums was compared. Meanwhile the components (IgG, IgA, IgM, IGF-I, INS, T3, T4) in calf serum were also tested after newborn calf fed the different seasonal colostrums, and the morbidity and mortality of newborn calf were statisted. Whether the effects on the acquired immunodeficiency of the newborn calf are significant were also discussed in this paper. The results are as follows:Compared with the agar double-diffusion, HPLC possess many advantages, such as quick analytical speed, high sensitivity and accuracy, high average recovery rate and good separation effect, and so on.The content of protein, IgG, butter-free milk solids, density and pH value in 168h, the content of total dry matter in 32h, the content of lactose in 64h-168h in bovine colostrums in summer were higher than that in winter; While the content of fat in 168h, lactose in 48h, the total dry matter in 48h-168h were lower than that in winter. The difference between the content of IgG in 2h, protein and fat in 32h, density in 64h were significant(P < 0.05), but the remaining were not significant(P > 0.05).The content of IgG, IgA, T3, T4, IGF-I, IgM, INS in the calf serum in 144h were higher than that in summer after newboth calf fed the colostrums in winter. The difference between IgG, T3, T4 were significant(P < 0.05), the difference between IgA, IgM, IGF-I and INS were not significant(P > 0.05). Different seasonal colostrums significantly influenced the passive immunization of the newborn calf(P < 0.05).
